目的:探讨β-连环蛋白(β-catenin)和细胞周期蛋白D1(CyclinD1)在人乳头瘤病毒(HPV)感染的子宫颈病变中的表达及临床病理意义。方法:采用免疫组化法和HybriMax导流杂交基因芯片技术,检测100例HPV感染的不同类型子宫颈病变组织中β-catenin和CyclinD1蛋白的表达,并以20例正常子宫颈组织作为正常对照组进行分析。结果:β-catenin在子宫颈鳞状细胞癌(CSCC)组中的异常表达率较低级别子宫颈上皮内瘤变(CIN-L)组及正常对照组有显著增高(P均<0.05);同时,高级别子宫颈上皮内瘤变(CIN-H)组的β-catenin异常表达率亦较CIN-L组及正常对照组有显著增高(P均<0.05);而CIN-L组与正常组间比较,CSCC组与CIN-H组间比较,β-catenin异常表达率差异均无统计学意义(P>0.05)。随着子宫颈病变严重程度的增加,CyclinD1的阳性表达率及染色强度呈递增趋势,在CIN和CSCC组织的细胞质及细胞核中β-catenin异常表达与CyclinD1表达呈正相关。结伦:β-catenin及CyclinD1蛋白表达异常与HPV感染的子宫颈病变密切相关。
Objective: To investigate the expression and clinical pathological significance of β-catenin and CyclinD1 in cervical lesions infected by human papillomavirus (HPV). Methods: The expressions of β-catenin and CyclinD1 in 100 cases of cervical lesions were detected by immunohistochemistry and HybriMax flow-through hybridization gene chip technique. Twenty normal cervical tissues were used as normal control group Analyze. Results: The abnormal expression rate of β-catenin in cervical squamous cell carcinoma (CSCC) group was significantly higher than that in CIN-L group and normal control group (all P <0.05). At the same time, the abnormal expression rate of β-catenin in CIN-H group was significantly higher than that in CIN-L group and normal control group (all P <0.05) There was no significant difference in the abnormal expression rates of β-catenin between CSCC group and CIN-H group (P> 0.05). As the severity of cervical lesions increased, the positive expression rate of CyclinD1 and the intensity of staining showed an increasing trend. The abnormal expression of CyclinD1 and β-catenin in the cytoplasm and nucleus of CIN and CSCC were positively correlated.
