目的:实验比较不同促进剂的溶液载体改变麝香酮透皮转运的促进增强作用。方法:将麝香酮分别溶于5组不同溶剂载体:①50%PG-EtOH、②50%PG-EtOH-0.5%LA、③50%PG-EtOH-5%Azone、④50%PG-EtOH-0.5%DA、⑤50%PG-生理盐水;采用 V-C扩散池,进行24 h的小鼠离体皮肤膜渗透实验,用GC法测定麝香酮膜渗透的累积累和渗透系统P,表达出麝香酮经皮促渗特征。结果:①、②组麝香酮渗透速率比⑤组分别提高 67倍和 95倍之多;含 Azone和 DA组分的载体24 h内的促渗穿透作用低于①和②组。结论:50%PG-EtOH体系是麝香酮经皮渗透的良好载体,LA对麝香酮有显著促渗作用;载体的化学性与麝香酮的渗透特性有相关性。
OBJECTIVE: To experimentally compare the carrier of different accelerants to change the enhancement of the transdermal transport of muskone. Method: Mussonone was dissolved in 5 different solvent carriers: 150% PG-EtOH, 250% PG-EtOH-0.5% LA, 350% PG-EtOH-5% Azone, and 450% PG-EtOH-0.5% DA. 550% PG-saline; using a VC diffusion cell for 24 hours in vitro skin membrane permeation test in mice, GC method was used to determine the permeation accumulation and infiltration system of musk ketone membrane, and to express the characteristics of percutaneous permeation of musk ketone. . RESULTS: The permeation rates of muskone in groups 1 and 2 were increased by 67-fold and 95-fold, respectively, compared with those in the 5 groups. The penetration-enhancing effects of carriers containing Azone and DA components within 24 h were lower than those in groups 1 and 2. CONCLUSION: The 50% PG-EtOH system is a good carrier for mucotone percutaneous penetration. LA has a significant effect on musk ketone penetration. The chemical nature of the carrier is related to the permeation characteristics of muskone.
