目的通过对白血病原代细胞的体外培养,研究白血病干细胞(LSC)的生物学特性。方法分离18例白血病患者骨髓或外周血中单个核细胞(MNC),在体外进行培养观察,并用流式细胞术方法检测分离的MNC中LSC的数量及和细胞表面标记。结果从白血病患者中分离的MNC在体外培养1周后基本死亡。流式检测分离的MNC中免疫表型是CD34+CD38-CD123+的细胞群约有0.676%(0.03%~2%)。结论白血病干细胞难以在体外培养生长及扩增。外周血及骨髓中分离白血病患者的单个核细胞中,极少部分细胞表达干细胞标志。
Objective To study the biological characteristics of leukemia stem cells (LSC) by culturing primary leukemia cells in vitro. Methods Mononuclear cells (MNCs) were isolated from bone marrow or peripheral blood of 18 patients with leukemia and cultured in vitro. The number of LSCs and cell surface markers in MNCs were detected by flow cytometry. Results MNC isolated from leukemia patients died essentially one week after in vitro culture. The immunophenotype of MNC isolated by flow cytometry was about 0.676% (0.03% -2%) of the population of CD34 + CD38-CD123 + cells. Conclusion Leukemia stem cells are difficult to culture in vitro growth and expansion. In peripheral blood and bone marrow mononuclear cells isolated from patients with leukemia, very few cells express stem cell markers.
