目的 研究青石棉纤维作用于A5 4 9细胞后IL 8蛋白和基因表达水平及酪氨酸激酶抑制剂PD980 5 9对IL 8表达的影响。方法 用定量RT PCR法测定青石棉诱导A5 4 9细胞后的IL 8mRNA水平 ;用免疫反应法测定A5 4 9细胞培养上清中IL 8蛋白水平。结果 不同浓度青石棉刺激A5 4 9细胞后 ,培养上清中IL- 8释放量明显增加 ,细胞中A5 4 9IL- 8mRNA表达显著上升 ,使用酪氨酸激酶抑制剂PD980 5 9可明显抑制IL- 8的蛋白及mRNA表达水平。结论 青石棉可诱导IL- 8的高表达 ,而这种表达可为PD980 5 9所抑制 ,提示IL -8的表达源于胞浆中MAPKs信号通路。
OBJECTIVE: To study the effects of crocidolite on IL-8 protein and gene expression in A549 cells and the effect of tyrosine kinase inhibitor PD98059 on IL-8 expression. Methods The level of IL 8 mRNA in A549 cells induced by crocidolite was detected by quantitative RT-PCR. The level of IL-8 in the culture supernatant of A549 cells was measured by immunoreactivity assay. Results After stimulation with different concentrations of crocidolite A549 cells, the release of IL-8 in the culture supernatant increased significantly, and the expression of A549-IL-8 mRNA in the cells increased significantly. PD98059, a tyrosine kinase inhibitor, 8 protein and mRNA expression levels. Conclusions Crocidolite could induce the high expression of IL-8, which was inhibited by PD98059, suggesting that the expression of IL-8 originated from the MAPKs signaling pathway in cytoplasm.
