目的:建立桑寄生药材的HPLC指纹图谱。方法:采用高效液相色谱法,用95%乙醇超声提取加水浴回流的混合提取方法制备样品,流动相:乙腈(A)∶(0.05 mol/L)KH2PO4-H3PO4缓冲溶液pH=3.0(B),线性洗脱梯度,洗脱程序如下:0~65 min乙腈10%→28%,65~85 min乙腈28%→52%。检测波长:220 nm;流速:1.0 mL/min。结果:分析了10批不同采摘时间的广东从化桑寄生药材,建立了桑寄生药材的HPLC指纹图谱,确立了16个共有峰,并且利用“中药色谱指纹图谱相似度评价软件”对不同样品之间的相似度进行计算。结论:该方法准确可靠,重复性好,为桑寄生药材的质量控制提供科学准确的依据。
Objective: To establish the HPLC fingerprint of medicinal material of Loranthaceae. Methods: The sample was prepared by high performance liquid chromatography with ultrasonic extraction of 95% ethanol and mixed with water. The mobile phase: acetonitrile (A): (0.05 mol/L) KH2PO4-H3PO4 buffer solution pH = 3.0 (B) The linear elution gradient elution procedure was as follows: 0-65 min acetonitrile 10%→28%, 65-85 min acetonitrile 28%→52%. Detection wavelength: 220 nm; flow rate: 1.0 mL/min. RESULTS: A total of 10 batches of medicinal herbs from different habitats in Guangdong Province were analyzed. HPLC fingerprints of medicinal herbs were established and 16 common peaks were established. The similarity evaluation software of traditional Chinese medicine chromatographic fingerprints was used to analyze different samples. The similarity between the calculations. Conclusion: The method is accurate, reliable, and reproducible. It provides scientific and accurate basis for the quality control of medicinal materials.
