目的 :探讨狗颅脑爆炸伤后脑组织中 TNF-α m RNA的变化及其规律。方法 :应用原位杂交技术分别检测狗颅脑爆炸伤后 30 min和 1、3、6 h大脑皮质、海马、下丘脑和脑干等不同脑区脑组织中 TNF-α m RNA的表达情况 ,并与正常对照组比较。应用电子计算机图像分析技术对其结果进行半定量分析。结果 :正常对照组狗脑组织内 TNF-α m RNA即有表达 ;颅脑爆炸伤后不同脑区脑组织中 TNF-α m RNA的表达量均明显升高 ,变化幅度以大脑皮质挫伤区和同侧海马最为显著。 TNF-αm RNA在伤后 30 m in表达量即明显增加 ,至伤后 1h达到高峰 ,伤后 3h呈下降趋势 ,但仍高于正常水平。伤侧大脑半球TNF- α m RNA的表达量明显高于对侧大脑半球。结论 :TNF- α m RNA在生理状态下即有表达 ,TNF- α参与正常的生理功能 ;颅脑爆炸伤后早期脑组织中 TNF- α的表达增多 ,提示 TNF- α可能参与了颅脑爆炸伤后的脑组织的病理改变
Objective: To investigate the changes and regularities of TNF-α m RNA in brain tissue after craniocerebral blast injury in dogs. Methods: The expression of TNF-αmRNA in cerebral cortex, hippocampus, hypothalamus and brain stem were detected by in situ hybridization at 30 min, 1, 3 and 6 h after TBI. And compared with the normal control group. The results were semi-quantitatively analyzed by computer image analysis technology. Results: The expression of TNF-α m RNA in the brain tissue of dogs in normal control group was significantly increased. The expression of TNF-α m RNA in brain tissue of different brain regions increased significantly in brain injury area Ipsilateral hippocampus most significant. The expression of TNF-αmRNA increased significantly 30 min after injury, reaching the peak at 1 h after injury and decreasing at 3 h but still higher than the normal level. The expression of TNF-αm RNA in the injured hemisphere was significantly higher than that in the contralateral hemisphere. CONCLUSION: TNF-αmRNA is expressed in physiological state and TNF-α is involved in normal physiological function. The expression of TNF-αin brain tissue increases after early brain injury, suggesting that TNF-α may be involved in brain-brain explosion Injured brain tissue pathological changes
