以库尔勒香梨幼叶为外植体,研究基本培养基种类、外源激素浓度、AgNO3浓度及接种叶片的放置方向对叶片不定芽再生的影响。结果表明:培养基的种类是影响叶片能否获得再生不定梢成功的关键,NN69培养基是香梨叶片再生不定芽的最佳培养基,诱导不定芽的分化以培养基NN69+TDZ 1.0mg/L+IBA 0.3mg/L为最佳;附加0.5mg/L AgNO3有利于叶片再生;以叶片远轴面接触培养基比近轴面接触培养基更利于不定芽的再生;结合21d暗培养香梨叶片再生频率最高可达到64.3%,再生芽数最高为2.59。
The young leaves of Korla fragrant pear were used as explants to study the effects of basic media, concentrations of exogenous hormones, AgNO3 concentration and orientation of inoculated leaves on adventitious bud regeneration. The results showed that: the type of medium was the key to the success of adventitious shoot regeneration. NN69 medium was the best medium for regeneration of adventitious buds of pear. The differentiation of adventitious buds was induced by medium NN69 + TDZ 1.0 mg / L + IBA 0.3mg / L is the best; Addition of 0.5mg / L AgNO3 is beneficial to leaf regeneration; Contact with the media near the leaf surface is more conducive to the regeneration of adventitious buds; Leaf regeneration frequency up to 64.3%, the highest number of regeneration buds 2.59.
