目的 :研究 3,7-二硝基二苯并溴五环盐对人慢性粒细胞红白血病细胞株K5 6 2增殖的影响及其凋亡机理。方法 :采用台盼蓝拒染法 ,MTT比色法及克隆形成率等方法 ,并用光学、荧光及电子显微镜观察照相 ,流式细胞术等方法分析DNA断裂的数值。结果 :cBr对K5 6 2具有杀伤作用 ,可明显抑制K5 6 2细胞增殖 ,呈现时间和剂量效应 :克隆形成率测得IC50 为 0 .4 97mmol/L ,流式细胞术测定亡率为 70 .34 % ;荧光显微镜和电镜观察到明显凋亡特征。结论 :cBr抑制肿瘤增殖是通过诱导肿瘤细胞凋亡实现的。
OBJECTIVE: To study the effect of 3,7-dinitrobenzofuran on the proliferation of human chronic myeloid leukemia cell line K562 and its apoptosis mechanism. Methods: The DNA fragmentation was analyzed by trypan blue exclusion, MTT colorimetric assay and clonogenic assay. The values of DNA fragmentation were analyzed by optical, fluorescence and electron microscopy, flow cytometry and other methods. Results: cBr could kill K562 cells and significantly inhibit the proliferation of K562 cells. The time and dose effects of cBr were observed. The IC50 of clone formation was 0.477mmol / L, and the apoptosis rate was 70% by flow cytometry. 34%; obvious apoptosis characteristics were observed by fluorescence microscope and electron microscope. Conclusion: cBr inhibits tumor proliferation by inducing tumor cell apoptosis.
