目的:研究小檗碱对高迁移率族蛋白B1(HMGB1)胞外释放的影响及其机制。方法:观察小檗碱对内毒素脂多糖激活后培养巨噬细胞株RAW264.7HMGB1基因表达和释放的作用,及对内毒素血症小鼠血清HMGB1水平和存活率的影响。结果:小檗碱(100μmol/L)明显抑制内毒素脂多糖激活后巨噬细胞HMGB1的mRNA表达和胞外释放(P<0.05)。内毒素血症小鼠腹腔注射小檗碱(5mg/kg)后,血清HMGB1水平降低,96h后小鼠存活率由28%提高至56%。结论:小檗碱具有抑制细胞HMGB1释放和保护内毒素血症小鼠作用。
Objective: To study the effect of berberine on the extracellular release of high mobility group box 1 (HMGB1) and its mechanism. Methods: To observe the effect of berberine on the expression and release of cultured macrophage RAW264.7HMGB1 after activation of endotoxin lipopolysaccharide, and the effect of berberine on serum HMGB1 level and survival rate in mice with endotoxemia. RESULTS: Berberine (100 μmol/L) significantly inhibited the mRNA expression and extracellular release of macrophage HMGB1 after lipopolysaccharide activation (P<0.05). After intraperitoneal injection of berberine (5 mg/kg) into mice with endotoxemia, serum HMGB1 levels decreased, and the survival rate of mice after 96 hours increased from 28% to 56%. CONCLUSION: Berberine has the effect of inhibiting the release of HMGB1 from cells and protecting endotoxemia in mice.
