对细脚拟青霉气升式发酵生产的胞外多糖(EPS)和胞内多糖(IPS)的单糖组成、氨基酸组成和分子量分别用气相色谱法、示差色谱法和氨基酸自动分析仪进行分析。结果表明:细脚拟青霉EPS和IPS各被分离为2种组分,分别命名为EPS F1,EPS F2,IPS F1和IPS F2;其中IPS F1的分子量最大,EPS F1和IPS F2分子量相同,EPS F2的分子量最小;这些纯化多糖组分的单糖成分差异很大,但它们的氨基酸成分基本相同。依次推测,多糖在胞内向胞外分泌时,其糖体会经过多种不同的修饰和加工,至使分泌出来的胞外多糖分子量比胞内多糖小,且糖体的组成成分出现较大变化。该结果为细脚拟青霉多糖不同组分的生理功能的深入研究及相关产品的开发提供一定的理论依据。
Monosaccharide composition, amino acid composition and molecular weight of extracellular polysaccharides (EPS) and intracellular polysaccharides (IPS) produced by gas-lift fermentation of Paecilomyces tenufaciens were analyzed by gas chromatography, differential spectroscopy and automatic amino acid analyzer respectively. The results showed that the EPS and IPS isolates were separated into two components named as EPS F1, EPS F2, IPS F1 and IPS F2 respectively. Among them, IPS F1 had the largest molecular weight and EPS F1 and IPS F2 had the same molecular weight, The molecular weight of EPS F2 is minimal; the monosaccharide components of these purified polysaccharide components vary widely, but their amino acid compositions are essentially the same. In turn speculated that the polysaccharide in the intracellular to extracellular secretion, the sugar through a variety of modifications and processing, to the secretion of exopolysaccharide molecular weight smaller than the intracellular polysaccharide, and the composition of the body of sugar showed a larger change. The results provide a theoretical basis for the further study of physiological functions of different components of Paecilomyces tenchii and the development of related products.
