目的:总结国内外关于细胞间紧密连接的破坏在乳腺癌浸润与转移中的作用。方法:应用Medline及CNKI期刊全文数据库系统以“乳腺癌、转移和紧密连接”为关键词检索1997-2010年的相关文献,分析文献30篇。纳入标准:1)乳腺癌的转移机制;2)紧密连接的结构及功能;3)紧密连接的破坏与乳腺癌转移的相关性。结果:紧密连接的破坏及紧密连接蛋白表达的下降可以促进EMT的发生,从而促进乳腺癌的浸润与转移。对于乳腺癌细胞TJ蛋白的研究可以进一步明确EMT在乳腺癌转移过程中确切机制,有助于掌握肿瘤演进过程,防治乳腺癌转移。结论:紧密连接的破坏与乳腺癌细胞的侵袭及转移存在一定的相关性。
OBJECTIVE: To summarize the role of cell-cell junctional destruction in infiltration and metastasis of breast cancer at home and abroad. Methods: Medline and CNKI journals full text database system was used to search the related literature of 1997-2010 with the key words of “Breast cancer, metastasis and tight junction”, and analyzed 30 articles. Inclusion criteria: 1) breast cancer metastasis mechanism; 2) the structure and function of tight junctions; and 3) the association of tight junction destruction with breast cancer metastasis. Results: The destruction of tight junctions and the decrease of tight junction protein expression can promote the occurrence of EMT and promote the infiltration and metastasis of breast cancer. The study of TJ protein in breast cancer cells can further clarify the exact mechanism of EMT in the process of breast cancer metastasis, help master the process of tumor evolution and prevent breast cancer metastasis. Conclusion: The damage of tight junctions is related to the invasion and metastasis of breast cancer cells.
