背景:从中药薏苡仁中提取具有抗癌活性的康莱特注射液,被广泛应用于肺癌的治疗。实验表明,对多种肿瘤株,包括LA795肺转移具有明显抑制作用。运用基因芯片技术,寻找康莱特小鼠肺腺癌LA795模型肺转移的差异表达基因。目的:应用基因芯片技术观察薏苡仁脂抗肿瘤转移反应性基因。设计:以实验动物为观察对象的对照实验。单位:中国中医研究院西苑医院肿瘤科。对象:实验于2003-02/08在中国中医研究院西苑医院实验室完成。接种LA795肺腺癌瘤株的T739小鼠,共40只,随机分为2组,每组20只。方法:以LA795肺腺癌小鼠肺转移组织,制备mRNA探针,应用含有4096条鼠基因的cDNA表达谱芯片,用ScanArray3000扫描仪扫描芯片荧光信号图像,分析差异表达的基因。主要观察指标:两组实验动物的差异表达基因。结果:发现在康莱特治疗组和空白对照组存在差异表达的基因,其中重复3次均差异表达的基因有27条,均表达增高的基因25条,表达降低的基因2条。其中12条是小鼠肌cDNA文库中的基因。经观察发现6条可能为与肿瘤的发生与发展及转移相关的基因。结论:运用基因表达谱芯片能够有效筛查出中药康莱特的相关差异基因。
BACKGROUND: Kanglaite injection with anticancer activity extracted from Chinese traditional medicine Coix is widely used in the treatment of lung cancer. Experiments show that a variety of tumor strains, including LA795 lung metastasis significantly inhibited. Using GeneChip technology to find differentially expressed genes in lung metastasis of lung adenocarcinoma model LA795 in Kanglaite mice. Objective: To observe the anti-tumor metastasis-responsive gene of Coix seed oil using gene chip technique. Design: A controlled experiment with experimental animals as observers. Unit: Xiyuan Hospital, China Academy of Traditional Chinese Medicine. PARTICIPANTS: The experiment was performed at the Laboratory of Xiyuan Hospital of China Academy of Traditional Chinese Medicine from February to August 2003. T739 mice vaccinated with LA795 lung adenocarcinoma were randomly divided into 2 groups with 20 mice in each group. Methods: Lung adenocarcinoma of LA795 mice were used for lung tissue transfection to prepare mRNA probes. CDNA microarray containing 4096 murine genes was used to scan the fluorescence signal of the chip with ScanArray3000 scanner to analyze differentially expressed genes. MAIN OUTCOME MEASURES: Differential expression of two experimental animals. RESULTS: There were 27 differentially expressed genes in KLT treated group and blank control group, of which 27 were differentially expressed in 3 replicates, with 25 highly expressed genes and 2 lowly expressed genes. Twelve of these genes are found in the mouse muscle cDNA library. It is observed that 6 genes may be related to the occurrence and development of tumor and metastasis. Conclusion: Gene expression profiling can effectively screen out the related genes of Kanglaite.
