目的:探讨抗癌生物活性肽作用于荷胃癌裸鼠后,对其肿瘤组织中凋亡相关蛋白p53表达变化的影响,探讨抗癌生物活性肽抑制胃癌细胞的作用机制。方法:将裸鼠接种胃癌MGC803细胞后使之致瘤。随机将裸鼠分为3组,对照组(NS组)11只、脾肽实验组(P组)12只、肝肽实验组(G组)11只,每天分别尾静脉注射给予生理盐水0.4 mL、脾肽0.4 mL(0.15mg/mL)、肝肽0.4 mL(0.05 mg/mL)。连续注射35d,停药4d后断头处死裸鼠,取肿瘤,以4%多聚甲醛固定,常规石蜡包埋,4μm厚连续切片。应用免疫组化SABC法检测突变型p53蛋白在实验组与对照组中的表达情况。结果:p53蛋白在NS组肿瘤中阳性表达较强,在P组、G组裸鼠肿瘤中p53蛋白的表达均比对照组的表达弱,且有显著性差异,(P<0.01)。结论:抗癌生物活性肽对MGC803荷瘤裸鼠有明显的抑瘤作用,其机制可能与抑制突变型p53蛋白的表达有关。
OBJECTIVE: To investigate the effect of anticancer bioactive peptide on the expression of p53 in gastric cancer tissue and to explore the mechanism of anticancer bioactive peptide inhibiting gastric cancer cells. Methods: Nude mice were inoculated with MGC803 gastric cancer cells after tumorigenicity. The nude mice were randomly divided into three groups: control group (n = 11), spleen peptide experimental group (group P) and hepatic peptide group (n = 11). The mice were injected with 0.4 mL normal saline 0.4 mL (0.15 mg / mL) of spleen peptide and 0.4 mL (0.05 mg / mL) of hepatic peptide. The rats were sacrificed 4 days after the continuous injection for 4 days. The nude mice were sacrificed and the tumors were removed. The tumors were fixed with 4% paraformaldehyde, embedded in paraffin and sliced in 4 μm thick. The expression of mutant p53 protein in experimental group and control group was detected by immunohistochemical SABC method. Results: The expression of p53 protein was significantly higher in NS group than in control group (P <0.01). The expression of p53 protein in group P and group G was weaker than that in control group (P <0.01). CONCLUSION: The anticancer bioactive peptide has obvious anti-tumor effect on MGC803 tumor-bearing nude mice, and its mechanism may be related to inhibiting the expression of mutant p53 protein.
