目的:探索不同激素组合对披针叶黄华愈伤组织诱导和增殖的影响。方法:以下胚轴为外植体,以MS为基本培养基,附加2,4-D、BA、NAA和IAA 4种植物激素,采用正交试验研究不同激素配比对披针叶黄华愈伤组织诱导和继代的影响。结果:愈伤组织诱导的最适培养基为MS+2,4-D(0.5 mg/L)+BA(1.0 mg/L)+NAA(1.0mg/L)+IAA(0.5 mg/L),各激素的影响程度为2,4-D>NAA>BA>IAA;愈伤组织继代的最适培养基为MS+2,4-D(0.1 mg/L)+BA(0.1 mg/L)+NAA(0.2 mg/L)+IAA(0.5 mg/L),各激素的影响程度为BA>2,4-D>IAA>NAA;愈伤组织在最优组合的继代培养基上的生长曲线呈“S”型。结论:首次成功建立披针叶黄华愈伤组织快速培养体系,为利用现代生物技术方法保护和利用披针叶黄华物种资源提供了一种新途径。
Objective: To explore the effects of different hormone combinations on the induction and proliferation of callus of Fagus sinica. METHODS: Four hypocotyls of 2,4-D, BA, NAA and IAA were used as explants. MS was used as the basic medium. Orthogonal test was used to study the effect of different hormones on callus Induction and the effects of subculture. Results: The optimum medium for callus induction was MS + 2,4-D (1.0 mg / L) + NAA (1.0 mg / L) The best medium for callus subculture was MS + 2,4-D (0.1 mg / L) + BA (0.1 mg / L) for 2,4-D> NAA> + NAA (0.2 mg / L) + IAA (0.5 mg / L), the effect of each hormone was BA> 2,4-D> IAA> NAA. The growth of callus on optimal combinations of subculture medium Curve was “S ” type. Conclusion: The rapid culture system of callus of Flos Licheni wort was successfully established for the first time, which provided a new way for using modern biotechnology methods to protect and utilize the resources of Phalaenopsis lanceolata.
