目的:探讨普罗布考(Probucol)对H_2O_2诱导的平滑肌细胞(SMCs)增殖的抑制作用。方法:采用细胞计数法和~3H-TdR掺入法观察 0. 1~100 μmol/L Probucol对新生牛血清(NCS)和过氧化氢(H_2O_2)促SMCs增殖的抑制作用。结果: 10μmol/L及100 μmol/L Probucol作用96 h,可抑制10%NCS刺激的SMCs增殖;而1,10及100 μmol/L Probucol作用72 h及96 h,均明显抑制 10μmol/L H_2O_2刺激的 SMCs增殖,且呈浓度依赖性(P<0. 05,p<0. 01)。 结论: Probucol抑制 NCS,H_2O_2刺激下体外培养的SMCs增殖,提示其对于动脉粥样硬化和经皮腔内冠状动脉成形术后再狭窄的防治可能具有应用前景。
AIM: To investigate the inhibitory effect of Probucol on H 2 O 2 -induced proliferation of smooth muscle cells (SMCs). Methods: Cell counting method and ~ 3H-TdR incorporation method was used to observe 0. Inhibition of Probucol by 1 ~ 100 μmol / L on Newborn Bovine Serum (NCS) and Hydrogen Peroxide (H_2O_2) - induced SMCs Proliferation. RESULTS: Probucol at 10 and 100 μmol / L for 96 h inhibited the proliferation of 10% NCS-stimulated SMCs. Probucol at 1, 10 and 100 μmol / L for 72 and 96 h inhibited the proliferation of SMMCs induced by 10 μmol / L H_2O_2 Of SMCs proliferated in a concentration-dependent manner (P <0.05, p <0.01). CONCLUSIONS: Probucol inhibits the proliferation of SMCs cultured in vitro stimulated by NCS and H 2 O 2, suggesting that it may have potential applications in the prevention and treatment of atherosclerosis and restenosis after percutaneous transluminal coronary angioplasty.
