目的:研究金银花和山银花的体外溶血作用,评价其潜在溶血风险。方法:以皂苷单体化合物、金银花有效部位及金银花、山银花药材等为实验材料,采用紫外分光光度法测定各类样品在不同浓度时对兔血红细胞的溶血率,绘制溶血率-质量浓度曲线。结果:忍冬苦苷A,忍冬苦苷B,灰毡毛忍冬皂苷甲,川续断皂苷乙和川续断皂苷Ⅵ在质量浓度≤1.0 g·L~(-1)时均未发生溶血。灰毡毛忍冬皂苷乙的溶血率随着质量浓度的升高而增大,当质量浓度≤0.6 g·L~(-1)时未发生溶血;当质量浓度为0.8 g·L~(-1)时,溶血率上升至11.7%;当质量浓度为1.0 g·L~(-1)时,溶血率达到50.4%。金银花各有效部位溶血作用的相对强弱为金银花总皂苷部位>总酚部位>总环烯醚萜部位。其中,金银花总环烯醚萜部位在质量浓度为0.1~1.2 g·L~(-1)时未发生溶血;金银花总酚部位质量浓度<1.0 g·L~(-1)时,未发生溶血。金银花总皂苷部位在质量浓度为0.6 g·L~(-1)时开始溶血,之后随着质量浓度的升高,其溶血率急剧增大,当质量浓度为1.2 g·L~(-1)时,溶血率55.3%。金银花野生品、栽培品与山银花的溶血率-质量浓度曲线较为相似,其溶血率急剧增大的质量浓度为2.0~4.0 g·L~(-1)。结论:金银花与山银花的溶血作用无显著性差异;各种皂苷单体化合物没有溶血作用或溶血作用较弱;金银花总皂苷部位的质量浓度为0.6 g·L~(-1)(相当于生药质量浓度20 g·L~(-1))时具有较弱的溶血作用。尽管如此,中药注射液一般不会达到如此高的药物浓度。因此,含有皂苷类成分的金银花与山银花可应用于中药注射剂,其潜在的溶血风险较小。
Objective: To study the in vitro hemolysis of honeysuckle and honeysuckle and evaluate the potential hemolysis risk. The relative strength of hemolysis of the effective parts of honeysuckle honeysuckle total saponin site> total phenolic> total iridoid site. Among them, the total iridoids of honeysuckle did not hemololyzed when the concentration of iridoid was 0.1-1.2 g · L -1, no hemolysis occurred when the total concentration of honeysuckle was less than 1.0 g · L -1 . When the concentration of total saponins of honeysuckle rose to 0.6 g · L -1, hemolysis started to increase rapidly with the increase of mass concentration. When the mass concentration was 1.2 g · L -1, Hemolysis rate was 55.3%. The hemolysis rate - mass concentration curves of wild honeysuckle cultivars, cultivars and honeysuckle flowers were similar, with the mass concentration of 2.0-4.0 g · L -1 increasing sharply. Conclusion: There is no significant difference in hemolysis between honeysuckle and Shanhain flower. The compounds of all saponins have no hemolysis or hemolysis. The concentration of total saponins in honeysuckle is 0.6 g · L -1 (equivalent to Crude drug concentration of 20 g · L -1) had weaker hemolysis. However, traditional Chinese medicine injections generally do not achieve such a high drug concentration. Therefore, the compositions containing saponins and honeysuckle can be applied to traditional Chinese medicine injections, the potential risk of hemolysis smaller.
