目的观察STZ诱导糖尿病大鼠胃平滑肌内质网应激相关蛋白GRP-78、CHOP、Caspase-12及其效应蛋白Bcl-2、Caspase-3的变化,探讨其在糖尿病胃轻瘫中发生的意义。方法根据STZ诱导6周大鼠出现糖尿病胃轻瘫症状及胃平滑肌细胞出现凋亡的前期实验结果,将SD大鼠分为正常对照组(NC)组和模型6周组(DM6W)。Western blot检测两组胃平滑肌GRP-78、CHOP、Caspase-12的表达,免疫组织化学染色检测两组胃平滑肌Bcl-2、Caspase-3的表达。结果 NC组胃平滑肌GRP-78、CHOP、Caspase-12的相对表达值低于DM6W组[(0.12±0.03)vs(0.23±0.03),(0.11±0.02)vs(0.24±0.03),(0.11±0.03)vs(0.24±0.04),P<0.01];NC组胃平滑肌Bcl-2的表达高于DM6W组[(491.91±22.96)vs(159.48±20.29),P<0.01];NC组胃平滑肌Caspase-3的表达低于DM6W组[(154.15±10.03)vs(489.45±52.08),P<0.01]。结论糖尿病胃轻瘫大鼠胃平滑肌存在内质网应激,并且参与诱导胃平滑细胞凋亡。
Objective To observe the changes of endoplasmic reticulum stress-related proteins GRP-78, CHOP, Caspase-12 and its effector proteins Bcl-2, Caspase-3 in STZ-induced diabetic rat gastric smooth muscle and to explore its significance in diabetic gastroparesis . Methods According to the preliminary experimental results of diabetic gastroparesis and apoptosis of gastric smooth muscle cells induced by STZ in 6 weeks, SD rats were divided into normal control group (NC) group and model 6 week group (DM6W). Western blot was used to detect the expression of GRP-78, CHOP and Caspase-12 in gastric smooth muscle of two groups. The expressions of Bcl-2 and Caspase-3 in gastric smooth muscle were detected by immunohistochemical staining. Results The relative expressions of GRP-78, CHOP and Caspase-12 in gastric smooth muscle in NC group were significantly lower than those in DM6W group [(0.12 ± 0.03) vs (0.23 ± 0.03), (0.11 ± 0.02) vs (0.24 ± 0.03) 0.03) vs (0.24 ± 0.04), P <0.01]. The expression of Bcl-2 in gastric smooth muscle in NC group was higher than that in DM6W group [(491.91 ± 22.96) vs (159.48 ± 20.29), P <0.01] -3 was lower than that in DM6W group [(154.15 ± 10.03) vs (489.45 ± 52.08), P <0.01]. Conclusion The gastric smooth muscle of diabetic rats with gastroparesis have endoplasmic reticulum stress and are involved in the induction of gastric smooth muscle cell apoptosis.
