目的 :为了验证 0 -(氯乙酰 -氨甲酰基 )烟曲霉醇 (TNP-470 )和顺铂 (DDP)联用的增效作用。方法 :将 64只 61 5小鼠腹腔内接种腹水瘤细胞株后 ,随机分为 4组 ,然后腹腔内分别注射生理盐水(第 1组 ) ,DDP(第 2组 ) ,TNP -470 (第 3组 )及联合用药 (第 4组 ) ,观察瘤细胞株对各组小鼠的腹水生成 ,瘤细胞数 ,存活率及生存时间。结果 :DDP,TNP -470及联合用药组与生理盐水对照组相比可显著抑制 61 5小鼠腹水的生成 ,减少瘤细胞数量 ,延长生存期 (P <0 .0 5 ) ;联合用药组与其它 3组比较在瘤细胞计数和瘤细胞存活率方面均有显著性差异 (P <0 .0 5 ) ;在小鼠存活率及生存时间方面 ,联合用药组延长最明显 ;在用药过程中 ,联合用药组可引起体重下降 ,但停止用药后体重又可逐渐得到恢复。结论 :TNP-470 ,DDP对小鼠腹水瘤腹腔种植后腹水的生成都有明显的抑制作用 ;TNP -470和DDP联用能起增效作用 ;体重下降这一副作用是可逆的
AIM: To demonstrate the synergistic effect of 0 - (chloroacetyl-carbamoyl) fumagillol (TNP-470) in combination with cisplatin (DDP). Methods: Sixty-four 61 5 mice were inoculated intraperitoneally with ascites tumor cell lines and then randomly divided into 4 groups. Then they were intraperitoneally injected with normal saline (group 1), DDP (group 2), TNP -470 (group 3 Group) and combination therapy (group 4). The ascites formation, tumor cell count, survival rate and survival time of tumor cells in each group were observed. Results: Compared with saline control group, DDP, TNP -470 and combination therapy group could significantly inhibit ascites formation, reduce the number of tumor cells and prolong the survival time of 61 5 mice (P <0.05) The other three groups showed significant difference in tumor cell count and tumor cell survival rate (P <0.05); in the survival rate of mice and survival time, the combination group was the most obvious extension; in the course of medication, Combination therapy group can cause weight loss, but after stopping the weight can be gradually restored. CONCLUSION: TNP-470 and DDP can significantly inhibit ascites production in mice after ascites tumor implantation. TNP-470 and DDP can increase synergism. The side effects of weight loss are reversible
