目的观察NIH稀毛小鼠在毛发发育周期中不同阶段的皮肤结构,并对小鼠的主要免疫指标进行检测。方法按C57BL/6小鼠毛发生长周期节点的时间观察NIH稀毛小鼠表型的增龄性改变,并剪取小鼠背部后侧皮肤,用4%多聚甲醛固定12 h,石蜡包埋切片,常规HE染色,光学显微镜观察皮肤中毛囊发育情况。选取6周龄的雌性小鼠,经小鼠眼眶静脉丛毛细管采血,血常规检测仪检测常用指标;经腹腔注射单增李斯特菌液,2×10~6 cfu/(100μl·只),感染48 h后,制备小鼠脾脏单细胞悬液,流式细胞术检测淋巴细胞比例。结果随着日龄的增长,NIH稀毛小鼠出现了无毛-稀毛-无毛的转变;在出生后第7天(P7)毛发结构异常,无法正常长出体表;P42时出现周期转换异常,皮下组织中仍存在部分结构异常的毛囊,至P180时未发生改变。NIH稀毛小鼠血液指标中红细胞数(RBC)、血红蛋白(HGB)、红细胞压积(HCT)等指标明显降低(P<0.05);淋巴细胞数及其在白细胞中的比例显著降低(P<0.05);中性粒细胞比例明显升高(P<0.05)。NIH稀毛小鼠感染单增李斯特杆菌前后脾脏中CD3~+CD4~+/CD3~+CD8~+比值均显著降低(P<0.05),CD3~+及CD19~+的比例差异无统计学意义(P>0.05)。结论 NIH稀毛小鼠毛发生长异常,生长过程中出现无毛-稀毛-无毛的表型改变,成年后会出现造血功能障碍和免疫力减弱。
Objective To observe the skin structure of NIH thin hairy mice at different stages of hair development cycle and to detect the main immune indexes of mice. Conclusion NIH hairless mice grow abnormally, and the phenotypes of hairless - hairless - hairless are changed during the growth. The hematopoietic dysfunction and the weakened immunity appear in adult NIH hairless mice.
