目的观察醋酸泼尼松对大鼠单位骨中骨矿、骨羟脯氨酸及对大鼠股骨密度和生物力学的影响,研究骨质量的改变与骨生物力学特性改变的联系。方法将20只SSD大鼠,随机分为正常对照组和激素组,每组10只。前者给蒸馏水5ml(/kg.d)灌胃,后者给醋酸泼尼松混悬液3.5mg(/5ml.kg.d)灌胃,88d后心脏抽血处死大鼠。检测单位骨中骨矿及骨羟脯氨酸含量;骨密度和骨生物力学检测。结果醋酸泼尼松抑制大鼠体重的增长,使单位骨中钙及羟脯氨酸含量减少。激素组股骨全段骨密度减小5.18%,股骨远端1cm处骨密度减小7.86%。醋酸泼尼松致股骨力学性能中的最大载荷减小10.78%,弹性载荷减小22.53%,弯曲能量减小24.94%,刚度系数减小18.03%。结论长期应用醋酸泼尼松可致骨基质和矿物质下降,骨密度降低,从而降低了骨的生物力学特性。
Objective To observe the effects of prednisone acetate on bone mineral density, hydroxyproline per unit bone area and the density and biomechanics of femur in rats, and to explore the relationship between the changes of bone mass and biomechanical properties. Methods Twenty SSD rats were randomly divided into normal control group and hormone group, with 10 rats in each group. The former was given 5ml distilled water (/kg.d) gavage, the latter to give prednisone acetate 3.5mg (/5ml.kg.d) gavage, 88d after cardiac arrest rats. Detection of bone mineral elements and bone hydroxyproline content; bone mineral density and bone biomechanical testing. Results Prednisone acetate inhibited the growth of body weight in rats and reduced the content of calcium and hydroxyproline in the unit bone. In the hormone group, the total femur bone mineral density decreased 5.18% and the bone density at the distal femur 1cm decreased 7.86%. The maximum load of femur induced by prednisone acetate decreased by 10.78%, the elastic load decreased by 22.53%, the bending energy decreased by 24.94% and the stiffness coefficient decreased by 18.03%. Conclusion Long-term use of prednisone acetate can lead to bone matrix and mineral decline, reduce bone mineral density, thereby reducing the biomechanical properties of bone.
