目的 探讨霍乱弧菌和其它致病弧菌分子遗传特征及相互关系。方法 用聚合酶链反应 (Polymerasechainreaction ;PCR)、DNA序列分析、随机扩增多态性DNA (RandomlyamplifiedpolymorphicDNA ;RAPD)和平均链锁聚类分析 ,对 3株O139群霍乱弧菌、3株O1群ElTor型、4株古典型和 3株副溶血等致病弧菌进行检测。结果 O139群霍乱弧菌和O1群含有相同的霍乱肠毒素 (CTX)A2 B亚单位基因 ,核苷酸序列同源性为 97 1% -98 9%。RAPD将不同弧菌分成 4类 ;即①O139群和ElTor型、②古典型、③副溶血和创伤弧菌及④河弧菌。O139群与O1群ElTor型DNA指纹图谱几乎完全一致 ,平均链锁距离为 0 ,与古典型相似 ,平均链锁距离为 2 0 7。与副溶血等致病弧菌差别较大 ,平均链锁距离为 6 76 - 8 5 4。结论 O139群霍乱弧菌与O1群ElTor型遗传特征相同 ,前者很可能由ElTor型进化而来。副溶血与创伤弧菌也具有相同遗传特征。霍乱弧菌和其它致病弧菌遗传特征表现出多态性。
Objective To investigate the molecular genetic characteristics and relationship of Vibrio cholerae and other pathogenic Vibrio spp. Methods Polymerase chain reaction (PCR), DNA sequence analysis, Randomly Amplified Polymorphic DNA (RAPD) and cluster-based average linkage analysis were used to analyze three strains of V. cholerae O139, three strains of O1 ElTor Type, 4 classical and 3 strains of hemolytic pathogenic Vibrio were detected. Results The V. cholerae O139 group and the O1 group contained the same cholera enterotoxin (CTX) A2 B subunit gene with a nucleotide sequence homology of 97 1% -98 9%. RAPD will be divided into four different Vibrio species; namely ①O139 group and ElTor type, ② classical, ③ Vibrio parahaemolyticus and Vibrio vulnificus and ④ River Vibrio. The O139 DNA fingerprinting of ElTor DNA was almost identical with that of O1 population with an average linkage distance of 0, similar to the classical one with an average linkage distance of 207. Vibrio parahaemolyticus and pathogenic bacteria differ greatly, with an average chain lock distance of 6 76 - 8 5 4. Conclusion The V. cholerae O139 strain has the same genetic characteristics as the ElTor type O1 strain, and the former is likely to evolve from the ElTor type. Vibrio parahaemolyticus and Vibrio vulnificus also have the same genetic characteristics. Vibrio cholerae and other pathogenic Vibrio genetic characteristics showed polymorphism.
