用电脉冲介导和DNA-磷酸钙共沉淀两种方法向缺乏胸腺嘧啶核苷激酶的小鼠LTK-细胞植入人HeLa S_3细胞DNA.转化后的小鼠细胞经HAT选择性培养,获得 TK~+表型的小鼠细胞.用~3H-TdR同位素掺入实验证实,转化后的小鼠细胞重新获得了TK基因编码的胸腺嘧啶核苷激酶.
The DNA of human HeLa S 3 cells was implanted into LTK cells lacking thymidine kinase by electric pulse mediated and DNA-calcium phosphate coprecipitation method. The transformed mouse cells were selectively cultured by HAT to obtain TK ~ + Phenotype of mouse cells with ~ 3H-TdR isotope incorporation experiments confirmed that the transformed mouse cells regain the TK gene encoding thymidine kinase.
