目的检测膀胱癌中生存素(Survivin)基因表达,探讨其与细胞凋亡的关系。方法采用原位杂交、免疫组织化学技术和DNA原位末端标记技术,检测46例膀胱癌组织、15例正常膀胱组织生存素mRNA及膀胱癌中caspase3蛋白和细胞凋亡。结果与正常膀胱组织相比,生存素mRNA显著表达于膀胱癌中,并与病理分级有关(P<0.05);膀胱癌中caspase3蛋白与凋亡细胞阳性率分别为76.1%和67.4%,凋亡指数均值为8.1%;生存素阴性组AI显著高于阳性组(P<0.01)。结论生存素选择性表达于膀胱癌中,通过抑制细胞凋亡,参与膀胱癌发生发展。
Objective To detect the expression of Survivin gene in bladder cancer and to explore its relationship with apoptosis. Methods 46 cases of bladder cancer, 15 cases of normal bladder tissue survivin mRNA and the expression of caspase3 protein and apoptosis in bladder cancer were detected by in situ hybridization, immunohistochemistry and DNA in situ end labeling. Results The expression of survivin mRNA in bladder cancer was significantly higher than that in normal bladder tissues (P <0.05). The positive rates of caspase 3 protein and apoptotic cells in bladder cancer were 76.1% and 67.4%, respectively The mean of index was 8.1%. The AI of survivin negative group was significantly higher than that of positive group (P <0.01). Conclusion Survivin is selectively expressed in bladder cancer and participates in the development of bladder cancer by inhibiting apoptosis.
