目的:观察洛伐他汀对大鼠心脏成纤维细胞多聚ADP核糖合成酶(PARP)-1活性及表达的影响。方法:采用PARP活性检测试剂盒和Western blot法检测洛伐他汀作用前后细胞内PARP-1的活性和蛋白表达水平。结果:使用洛伐他汀后,PARP-1的活性和蛋白表达均下调;再加用甲羟戊酸后,PARP-1的活性和表达下调被逆转。结论:洛伐他汀不仅可抑制血管紧张素Ⅱ刺激的心脏成纤维细胞PARP-1的活性及表达,还可在基础状态下抑制PARP-1的活性及表达,以上抑制作用都可以被甲羟戊酸逆转。
Objective: To observe the effect of lovastatin on the activity and expression of poly ADP ribose synthase (PARP) -1 in rat cardiac fibroblasts. Methods: PARP activity assay kit and Western blot were used to detect the activity of PARP-1 and protein expression level before and after lovastatin treatment. Results: After lovastatin treatment, the activity and protein expression of PARP-1 were down-regulated. After the addition of mevalonate, the activity of PARP-1 and the down-regulation of PARP-1 were reversed. Conclusion: Lovastatin can not only inhibit the activity and expression of PARP-1 in cardiac fibroblasts stimulated by angiotensin Ⅱ, but also inhibit the activity and expression of PARP-1 in basal state, Acid reversed.
