黄芪注射液拮抗柴油机尾气颗粒物诱导的小鼠生殖功能改变

目的研究黄芪注射液对柴油机尾气颗粒物诱导的小鼠生殖功能改变的影响。方法5周龄雄性ICR小鼠60只随机分为对照PBS组,DEP染毒组(12、3、0.8μg/μl),黄芪注射液干预组(10mg/ml、20mg/ml)。染毒模型采用De Hennezel法。对照组、染毒组每日腹腔注射0.9%生理盐水1ml,干预组每日分别腹腔注射10mg/ml、20mg/ml的黄芪注射液1ml共28d,每周2次染毒连续4周,第6周处死。检测体重、睾丸、附睾脏器系数,精子密度、总活力,体外受精率。结果各组间睾丸重量、睾丸系数无统计学差异(P>0.05)。3、12μg/μl DEP染毒组与PBS组、0.8μg/μ1DEP染毒组比较附睾重量、附睾系数,精子密度、总活力、体外受精率均有统计学意义(P<0.05);3、12μg/μl DEP的黄芪注射液干预组与对应DEP染毒组比较附睾重量、附睾系数、精子密度、总活力、受精率有显著差异(P<0.05)但不存在剂量依赖关系(P>0.05)。总活力和体外受精率之间呈正相关(P=0.000)。结论每日连续运用10mg/ml、20mg/ ml的黄芪注射液1ml可以拮抗由柴油机尾气颗粒物诱导的雄性ICR小鼠生殖功能的改变。 Objective To study the effect of Huangqi injection on the changes of reproductive function in mice induced by diesel exhaust particles. Methods A total of 60 5-week-old male ICR mice were randomly divided into control PBS group, DEP treated group (12, 3, and 0.8 μg/μl) and Huangqi injection intervention group (10 mg/ml, 20 mg/ml). The De Hennezel method was used for the exposure model. The control group and the poisoned group were intraperitoneally injected with 1 ml of 0.9% physiological saline daily. The intervention group was intraperitoneally injected with 10 ml/ml and 20 mg/ml of Astragalus membranaceus injection for a total of 28 days each day, and was exposed twice a week for 4 weeks. Zhou was executed. Weight, testis, epididymal organ coefficient, sperm density, total vitality, in vitro fertilization rate were measured. Results There was no significant difference in testis weight and testis coefficient between the groups (P>0.05). 3, 12μg/μl DEP poisoned group and PBS group, 0.8μg/μ1DEP poisoned group compared epididymis weight, epididymis coefficient, sperm density, total viability, in vitro fertilization rate were statistically significant (P <0.05); 3, 12μg Compared with the DEP-treated group, the epididymis weight, epididymis index, sperm density, total viability, and fertilization rate were significantly different (P<0.05) but there was no dose-dependent relationship (P>0.05). There was a positive correlation between total viability and in vitro fertilization rate (P=0.000). Conclusion The continuous application of 10 ml/ml and 20 mg/ml Astragalus injection 1 ml can antagonize the changes of reproductive function of male ICR mice induced by diesel exhaust particles.
