目的探讨聚合胶原对肌成纤维细胞表型转化的促进作用及其可能机制。方法用聚合胶原培养小鼠成纤维细胞,观察聚合胶原对成纤维细胞形态学及生长曲线影响。蛋白印迹法分析PTEN及Akt Ser473及p42/44磷酸化水平的情况。结果在聚合胶原中培养的成纤维细胞可以向肌成纤维细胞转化。在聚合胶原的诱导下,成纤维细胞内PTEN含量减少,磷酸化Akt增多,同时细胞内磷酸化p42/44MAPK含量增多。结论聚合胶原可以诱导成纤维细胞向肌成纤维细胞转化。其机制可能与PTEN和p42/44MAPK信号通路相关。
Objective To investigate the effect of polymerized collagen on the phenotype transformation of myofibroblasts and its possible mechanism. Methods The mouse fibroblasts were cultured with collagen and the effects of collagen on the morphology and growth curve of fibroblasts were observed. Western blot analysis of PTEN and Akt Ser473 and p42 / 44 phosphorylation levels. As a result, fibroblasts cultured in polymerized collagen can be transformed into myofibroblasts. Under the condition of polymerized collagen, the content of PTEN in fibroblasts decreased, the phosphorylated Akt increased and the content of intracellular phosphorylated p42 / 44MAPK increased. Conclusion Polymeric collagen induces the transformation of fibroblasts into myofibroblasts. Its mechanism may be related to PTEN and p42 / 44 MAPK signaling pathway.
