目的:研究茉莉酸甲酯(methyl jasmonate,MJ)对曼陀罗毛状根中主要次生代谢产物东莨菪碱和莨菪碱成分的积累和向培养基中的释放的影响。方法:1/2 MS液体培养C58C1农杆菌诱导的曼陀罗毛状根培养18 d后,液体培养基中加入化学诱导子———茉莉酸甲酯,采用HPLC测定MJ处理后不同浓度梯度及不同时间段(0,3,6,9,12 d)曼陀罗毛状根中东莨菪碱和莨菪碱的含量以及其在培养基中的含量。结果:曼陀罗毛状根经MJ处理后3,6,9,12 d的东莨菪碱的质量分数分别达0.419,0.439,0.431,0.374 mg.g-1,分别是同时期未经MJ处理的1.36,1.42,1.17,1.12倍;莨菪碱的质量分数达1.493,0.817,0.723,0.698 mg.g-1,分别是同时期未经MJ处理曼陀罗毛状根的2.28,1.11,0.63,0.70倍。结论:MJ能显著促进曼陀罗毛状根中东莨菪碱成分的积累并向培养基中释放;MJ能显著促进处理3 d和6 d后曼陀罗毛状根中莨菪碱成分的积累并向培养基中释放。
Objective: To investigate the effects of methyl jasmonate (MJ) on the accumulation of scopolamine and hyoscyamine in the hairy roots of Datura stramonium and its release into culture medium. Methods: Cultured mandibular hairy roots induced by Agrobacterium tumefaciens C18C1 were cultured in 1/2 MS medium for 18 days. Methyl jasmonate, a chemical inducer, was added to the liquid medium. Different concentrations of MJ were measured by HPLC. The content of scopolamine and hyoscyamine in the hairy roots of Datura stramoner at different time periods (0, 3, 6, 9, 12 d) and their contents in the culture medium. Results: The mass fractions of scopolamine on the 3rd, 6th, 9th, 12th and 12th days of MJ treated by MJ were 0.419, 0.439, 0.431 and 0.374 mg.g-1, respectively, which were respectively 1.36 , 1.42,1.17 and 1.12 times respectively. The mass fraction of hyoscyamine was 1.493,0.817,0.723,0.698 mg.g-1, which was 2.28,1.11,0.63,0.70 times higher than that of the untreated MJ . CONCLUSION: MJ can significantly promote the accumulation of scopolamine in the hairy roots of Datura stramonium and release it to the medium. MJ can significantly promote the accumulation of scopolamine in the hairy roots of Datura stramon after 3 d and 6 d, Base release.
