目的:探讨可溶性Jagged-1/Fc嵌合蛋白对小鼠淋巴细胞产生细胞因子的影响。方法:不同时间和不同浓度Jagged-1/Fc处理小鼠淋巴细胞,ELISA检测其培养上清中细胞因子的水平,并用Westernblot分析相关蛋白NICD和Del-tex的表达。结果:低浓度Jagged-1/Fc对淋巴细胞生成IL-17无明显影响,但高浓度Jagged-1/Fc(500μg/L)组IL-17的表达量是对照组的64.75倍,并且随着时间的增加而减少;IFN-γ随着Jagged-1/Fc浓度的增加而减少。Westernblot分析显示Jagged-1/Fc处理组的NICD和Deltex的表达明显高于对照组和Anti-Jagged-1组。结论:高浓度Jagged-1/Fc能够诱导淋巴细胞表达IL-17,可能在指导初始T细胞向Th17细胞偏离中发挥作用。
AIM: To investigate the effect of soluble Jagged-1 / Fc chimeric protein on cytokines production in mouse lymphocytes. Methods: Mice lymphocytes were treated with Jagged-1 / Fc at different time and different concentrations. The levels of cytokines in the culture supernatants were detected by ELISA. The expression of NICD and Del-tex was analyzed by Western blot. Results: Low concentration of Jagged-1 / Fc had no effect on IL-17 production in lymphocytes, but IL-17 expression in Jagged-1 / Fc 500μg / L group was 64.75 times higher than that in control group Time decreased; IFN-γ decreased with the increase of Jagged-1 / Fc concentration. Western blot analysis showed that the expression of NICD and Deltex in Jagged-1 / Fc group was significantly higher than that in control group and Anti-Jagged-1 group. Conclusion: High concentration of Jagged-1 / Fc can induce the expression of IL-17 in lymphocytes, which may play a role in guiding the initial T cells to deviate from Th17 cells.
