目的研究复方鸦胆子油脂质体冻干粉的性质,建立其质量评价方法。方法考察复方鸦胆子油脂质体的形态、粒径分布、Zeta电位,采用TLC法对其定性鉴别,并采用溶剂萃取法测定脂质体包封率。同时,通过UV和GC的方法测定其有效成分的含量。结果复方鸦胆子油脂质体冻干粉色泽均匀,质地细腻,形态饱满,其平均粒径为170.3 nm,多分散系数(PDI)为0.258,Zeta电位为-32.1 mV,带负电,脂质体pH为5.05。TLC鉴别中检出蟾酥、鸦胆子油斑点清晰可见。脂质体冻干粉的平均包封率为91.90%,吲哚类生物碱的平均质量分数为5.19 mg/g,油酸的平均质量分数为79.08 mg/g,亚油酸的平均质量分数50.41 mg/g。脂质体的平均过氧化值为0.030 5。结论上述方法适用于复方鸦胆子油脂质体冻干粉的质量评价,可有效反映其各方面的性质。
Objective To study the properties of the compound Brucea javanica oil liposome lyophilized powder and establish its quality evaluation method. Methods The morphology, particle size distribution and zeta potential of compound Brucea javanica oil were studied qualitatively by TLC, and the encapsulation efficiency was determined by solvent extraction. At the same time, the contents of active ingredients were determined by UV and GC methods. Results The results showed that the average volume of the lyophilized powder was 170.3 nm, the polydispersity (PDI) was 0.258 and the Zeta potential was -32.1 mV, with negative charge and liposome pH Is 5.05. TLC identification identified toad, Brucea javanica oil spots clearly visible. The average entrapment efficiency of liposomes was 91.90%, the average mass fraction of indole alkaloids was 5.19 mg / g, the average content of oleic acid was 79.08 mg / g, the average content of linoleic acid was 50.41 mg / g. The liposomes had an average peroxide value of 0.0305. Conclusion The above method is suitable for the quality evaluation of the compound Brucea javanica oil liposome lyophilized powder, which can effectively reflect the properties of all aspects.
