本文报告了以打点杂交法研究c-abl癌基因在人CML K562细胞株中扩增的结果。将α-~(32)P标记v-abl探针与经系列稀释并点加在硝酸纤维膜上的DNA进行杂交。可见K562细胞DNA信号较正常人DNA者明显增强,表明K562细胞中c-abl癌基因有特异性扩增作用。提示c-abl癌基因扩增可能与CML的发病机理有密切关系。
This paper reports the results of a dot blot hybridization study of c-abl oncogene amplification in human CML K562 cell line. The α-32 P-labeled v-abl probe was hybridized with serially diluted DNA spotted on nitrocellulose membrane. It can be seen that the DNA signal of K562 cells is significantly enhanced compared with that of normal human DNA, indicating that the c-abl oncogene has a specific amplification in K562 cells. Tip c-abl oncogene amplification may be closely related to the pathogenesis of CML.
