目的观察薏苡仁多糖对小鼠的抗氧化作用,并探讨其机制。方法以ICR清洁级小鼠为研究对象,腹腔注射CCl4造模。以VE为阳性对照,给药组[薏苡仁纯多糖低、中、高剂量(250、500、1000mg/kg)]和薏苡仁粗多糖低、中、高剂量(250、500、1000mg/kg)分别先经灌胃28d进行预防。以全血SOD与GSH-Px活性、GSH浓度、血清总抗氧化能力(T-AOC)反映抗氧化功能;观察肝脏GOT、GPT活力单位、丙二醛(MDA)含量和肝脏指数变化。结果与模型组相比,薏苡仁多糖可显著增强小鼠SOD、GSH-Px活性,有效防止CCl4对肝脏组织的伤害,明显抑制肝细胞GOT、GPT活性与MDA含量的异常上升,并呈现明显的量-效关系,且以薏苡仁纯多糖的效果为好。结论薏苡仁多糖能显著增强小鼠的抗氧化功能,有效解除CCl4肝中毒效应,其机制可能与提高机体的抗氧化酶活性和GSH浓度,对抗脂质过氧化反应有关。
Objective To observe the anti-oxidation effects of Coix Cipa polysaccharide on mice and to explore its mechanism. Methods ICR clean grade mice were used as subjects, and CCl4 was injected intraperitoneally. With VE as positive control, low, medium, and high doses of Coix Seed polysaccharide (250, 500, 1000 mg/kg) and low, medium, and high doses of Coix Seed polysaccharide (250, 500, 1000 mg/kg) They were pretreated with gavage for 28 days. Antioxidation function was reflected by whole blood SOD and GSH-Px activity, GSH concentration, and serum total antioxidant capacity (T-AOC); changes in liver GOT, GPT activity units, malondialdehyde (MDA) content and liver index were observed. Results Compared with the model group, Coix seed polysaccharide can significantly increase the activity of SOD and GSH-Px in mice, effectively prevent the damage of CCl4 to liver tissue, and significantly inhibit the abnormal increase of GOT, GPT activity and MDA content in hepatocytes, and show obvious The quantity-effect relationship is good, and the effect of pure polysaccharides is preferred. Conclusion Coix seed polysaccharide can significantly enhance the antioxidation function of mice and effectively relieve the liver poisoning effect of CCl4. The mechanism may be related to the increase of antioxidant enzyme activity and GSH concentration of the body, and the anti-lipid peroxidation reaction.
