目的:通过检测成人白血病中细胞因子信号转导抑制因子-1(SOCS-1)基因的表达水平,研究其在白血病发病中的作用。方法:运用实时定量PCR(Real-time PCR)方法对28例白血病患者和3株白血病细胞株(Jurkat、Raji、U937),进行SOCS-1基因表达情况的研究,并以10例健康人为正常对照组。结果:28例白血病患者(急性白血病18例,慢性白血病10例)的SOCS-1基因表达量较正常对照组明显降低(P<0.01);其中急性白血病组与正常对照组相比较,SOCS-1基因表达水平显著降低(P<0.01);而慢性白血病组的SOCS-1基因表达水平与正常对照组相比较,差异无统计学意义(P>0.05)。3株白血病细胞株与正常对照组比较,SOCS-1基因表达水平也显著降低(P<0.01)。急性白血病患者的SOCS-1基因表达量在患者的不同年龄、性别和病程阶段差异均无统计学意义(P>0.05)。结论:SOCS-1基因在成人急性白血病中表达水平明显降低(P<0.01),提示SOCS-1基因在急性白血病的发生发展中可能具有一定作用。
OBJECTIVE: To investigate the role of SOCS-1 gene in the pathogenesis of leukemia by detecting the expression of cytokine signaling suppressor-1 (SOCS-1) gene in adult leukemia. Methods: The expression of SOCS-1 gene in 28 leukemia patients and 3 leukemia cell lines (Jurkat, Raji, U937) were studied by Real-time PCR. Ten healthy individuals were selected as normal control group. Results: The expression of SOCS-1 in 28 leukemia patients (18 cases of acute leukemia and 10 cases of chronic leukemia) was significantly lower than that of the normal control group (P <0.01). Compared with the normal control group, SOCS-1 (P <0.01). However, the expression of SOCS-1 gene in chronic leukemia group was not significantly different from that in normal control group (P> 0.05). SOCS-1 gene expression was also significantly decreased in 3 leukemia cell lines compared with the normal control group (P <0.01). There was no significant difference in SOCS-1 gene expression in patients with acute leukemia at different age, sex and disease stage (P> 0.05). CONCLUSIONS: The expression of SOCS-1 gene in adult acute leukemia was significantly lower (P <0.01), suggesting SOCS-1 gene may play a role in the development of acute leukemia.
