目的 :建立测定人血浆中司巴沙星药物浓度的 RP- HPL C法 ,为临床合理用药提供必要的分析方法。方法 :采用HPL C法 ,色谱柱为 YWG RP- C1 8( 15 0 mm× 4.6 m m,10 μm) ;流动相为 5 0 m mol/L Na Ac- HAc缓冲液 ( p H5 .0 )∶乙腈∶甲醇 =70∶ 2 0∶ 10 (含 0 .0 4%三乙胺 ,调 p H2 .40 ) ,检测波长 2 99nm。血样 (含内标对羟基苯乙酮 )用 7ml二氯甲烷一次提取 ,浓缩后取 2 0 μl进样作 HPL C分析。结果 :线性方程为 Y=2 .5 5 6 c- 0 .1398( r=0 .9991,n=8) ,最低检测浓度为 0 .1μg/m l,方法回收率为 10 0 .5 4% ,精密度 RSD<4%。 36名性病患者 po司巴沙星片 2 0 0 m g,12 h后血药浓度范围为 0 .38~ 1.80 μg/ml。结论 :本方法准确、简便、灵敏 ,可用于测定该药在人血浆中的药物浓度
OBJECTIVE: To establish a RP-HPL C method for the determination of concentration of sparfloxacin in human plasma and provide the necessary analytical methods for clinical rational drug use. Methods: The HPL C method was used. The chromatographic column was YWG RP-C1 8 (150 mm × 4.6 mm, 10 μm). The mobile phase consisted of 50 mM Na-HAc buffer (p H5 .0) : Methanol = 70: 2 0:10 (containing 0.04% triethylamine, adjusted p H2 .40), the detection wavelength of 299nm. Blood samples (containing the internal standard of hydroxyacetophenone) with 7ml of dichloromethane extracted once, after concentration 20Ol injection for HPL C analysis. Results: The linear equation was Y = 2.556 c-0.1398 (r = 0.9991, n = 8), the lowest detection concentration was 0.1μg / ml, the recovery rate was 100.44% Precision RSD <4%. Thirty-six STD patients received poibacacin 200 mg daily for 12 h, and the plasma concentration ranged from 0.38 to 1.80 μg / ml. Conclusion: The method is accurate, simple and sensitive and can be used to determine the drug concentration of the drug in human plasma
