目的 :研究Ku70和Ku80基因在系统性红斑狼疮 (systemiclupuserythematosus,SLE)患者皮肤表达的变化及其与SLE发病的关系。方法 :利用Ku70、Ku80地高辛标记的cDNAs探针对皮损区、非皮损区及正常对照皮肤进行原位杂交。结果 :Ku70、Ku80mRNAs的表达量从高到低依次为SLE皮损区、SLE非皮损区、正常皮肤。另外还发现在皮肤的层次上真皮浅层单一核细胞Ku70、Ku80mRNAs表达量高于表皮基底层细胞、表皮棘细胞层和颗粒层细胞。结论 :Ku70、Ku80mRNAs的表达在SLE患者皮肤上发病区高于非发病区 ,更大于正常皮肤。Ku70、Ku80mRNAs表达的变化与SLE的发病有关 ,其机制有待进一步研究
Objective: To study the changes of Ku70 and Ku80 gene expression in the skin of patients with systemic lupus erythematosus (SLE) and their relationship with the pathogenesis of SLE. Methods: Ku70 and Ku80 digoxigenin-labeled cDNAs probes were used to in situ hybridize the skin lesions, non-skin lesions and normal controls. Results: The expressions of Ku70 and Ku80 mRNA were highest in SLE lesions, SLE non-lesions and normal skin. In addition, it was found that the expression of Ku70 and Ku80 mRNA in dermal superficial layers of mononuclear cells at the level of skin was higher than that of epidermal basal cells, epidermal spine and granulosa cells. Conclusion: The expression of Ku70 and Ku80 mRNA in the skin of patients with SLE is higher than that of non-affected area, more than normal skin. Ku70, Ku80 mRNA expression changes and the pathogenesis of SLE, the mechanism needs further study
