目的:探讨复方虎杖提取物对高脂饲料致高脂血症模型大鼠血液流变学的作用。方法:采用高脂饲料喂养SD大鼠连续11周建立高脂血症模型,第3周取血测定血脂水平判断模型是否复制成功,模型复制成功后连续灌胃给予受试药(复方虎杖提取物)3个剂量组(4g/kg、8g/kg、12g/kg)及阳性药组(辛伐他汀片,4mg/kg)8周。末次给药后大鼠下腔静脉取血,加肝素钠抗凝,测血液流变学指标。结果:末次给药后,与正常对照组比较,模型对照组大鼠全血黏度中黏度200/s、黏度30/s、黏度3/s、黏度150/s、黏度50/s、黏度1/s、高切还原黏度、全血高切流阻、全血中切流阻、全血低切流阻、红细胞压积及卡松黏度均显著升高(P<0.01,P<0.05)。与模型对照组比较:①复方虎杖提取物低剂量(4g/kg)组全血黏度中黏度3/s、全血低切流阻、卡松屈服应力显著降低(P<0.01),黏度200/s、黏度3/s、黏度150/s、黏度50/s、黏度1/s、血浆黏度、全血高切流阻、全血低切流阻、血沉方程K值、卡松黏度明显降低(P<0.05);②中剂量(8g/kg)组全血黏度中黏度3/s、黏度1/s、全血低切流阻、卡松屈服应力显著降低(P<0.01),黏度200/s、黏度30/s、黏度150/s、黏度50/s、血浆黏度、高切还原黏度、全血高切流阻、全血中切流阻、血沉方程K值、红细胞压积、明显降低(P<0.05),红细胞变形指数明显升高(P<0.05);③高剂量(12g/kg)组全血黏度中黏度3/s、血浆黏度、全血低切流阻、卡松屈服应力、红细胞压积显著降低(P<0.01),黏度200/s、黏度150/s、黏度50/s、黏度30/s、血沉方程K值降低(P<0.05),红细胞变形指数显著升高(P<0.01)。结论:复方虎杖提取物4g/kg、8g/kg、12g/kg三个剂量组可以改善高脂饲料致高脂血症模型大鼠的血液黏滞性。
Objective: To investigate the effects of compound Polygonum cuspidatum extract on hemorrheology in hyperlipidemic rats induced by high fat diet. Methods: SD rats were fed with high-fat diet for 11 consecutive weeks to establish hyperlipidemia model. Blood lipid level was determined in the third week to determine if the model was successfully replicated. After the model was successfully replicated, the test drug was given to the SD rats (Compound Polygonum cuspidatum extract ) For 3 weeks (4g / kg, 8g / kg, 12g / kg) and positive group (simvastatin tablets, 4mg / kg) for 8 weeks. After the last administration of rat vena cava blood collection, plus heparin anticoagulation, measuring blood rheology indicators. Results: After the last administration, the whole blood viscosity of model control group was 200 / s, viscosity 30 / s, viscosity 3 / s, viscosity 150 / s, viscosity 50 / s and viscosity 1 / (P <0.01, P <0.05). The viscosity of high shear reduction, high shear resistance of whole blood, whole blood shear flow resistance, whole blood low shear flow resistance, hematocrit and cardon viscosity were all significantly increased. Compared with the model control group: ①Compared with the model control group, the viscosity of the whole blood in the low dose (4g / kg) group was significantly lower (P <0.01) and the viscosity of 200 / Viscosity 150 / s, viscosity 50 / s, viscosity 1 / s, plasma viscosity, whole blood high shear resistance, whole blood low shear resistance, erythrocyte sedimentation equation K value, (P <0.05); ② The middle viscosity (8g / kg) group showed a significant decrease in the whole blood viscosity of viscosity 3 / s, viscosity 1 / s, s, viscosity 30 / s, viscosity 150 / s, viscosity 50 / s, plasma viscosity, high shear reduction viscosity, whole blood high shear resistance, whole blood shear flow resistance, erythrocyte sedimentation rate K value, hematocrit, (P <0.05), and the index of erythrocyte deformation was significantly increased (P <0.05). ③ The viscosity of whole blood at high dose (12g / kg) , The hematocrit decreased significantly (P <0.01), the viscosity of 200 / s, the viscosity of 150 / s, the viscosity of 50 / s, the viscosity of 30 / s and the erythrocyte sedimentation index K decreased (P < P <0.01). CONCLUSION: The three doses of compound Polygonum cuspidatum extract 4g / kg, 8g / kg and 12g / kg can improve the blood viscosity of hyperlipemia model rats induced by high-fat diet.
