目的:探讨健脾清化方对2型糖尿病大鼠(T2DM)肝脏Ras蛋白特异鸟嘌呤核苷酸释放因子1(RASGRP1)的影响。方法:采用高脂饮食联合链脲佐菌素(STZ)建立稳定的T2DM大鼠模型,随机分为对照组、模型组和健脾清化方组,连续灌胃给药4周,观察大鼠空腹血糖(FBG)和糖化血红蛋白(Hb A1C)水平变化,RT-PCR法和Western blot法分别检测大鼠肝脏组织RASGRP1基因和蛋白的表达水平。结果:与模型组比较,健脾清化方下调T2DM大鼠的FBG和Hb A1C水平(P<0.01),以及肝脏组织中RASGRP1基因和蛋白的表达(P<0.01)。结论:健脾清化方能够降低T2DM大鼠的FBG和Hb A1C水平,改善胰岛细胞功能,其作用机制可能与下调肝脏组织中RASGRP1的表达有关。
Objective: To investigate the effects of Jianpi Qinghua Recipe (LQJ) on Ras protein specific guanine nucleotide release factor 1 (RASGRP1) in type 2 diabetic rats (T2DM). Methods: A stable T2DM rat model was established by high-fat diet combined with streptozotocin (STZ). The rats were randomly divided into control group, model group and Jianpi Qinghua Fang group. After continuous gavage for 4 weeks, Fasting blood glucose (FBG) and Hb A1C levels were detected by RT-PCR and Western blot. The expression of RASGRP1 gene and protein were detected by RT-PCR and Western blot respectively. Results: Compared with the model group, Jianpi Qinghua Decoction down-regulated the FBG and Hb A1C levels (P <0.01) and the expression of RASGRP1 gene and protein in the liver of T2DM rats (P <0.01). Conclusion: Jianpi Qinghua Decoction can reduce the levels of FBG and Hb A1C and improve the function of islet cells in T2DM rats. The mechanism may be related to the down-regulation of RASGRP1 expression in liver tissues.
