为了探索一种新的中性鞘糖脂 (neutral glycosphingolipids,N- GSL s)单一组分的纯化分离鉴定方法 ,将通过改良的 Hakomori方法提取的牛脑中性鞘糖脂经过加压柱层析 ,梯度甲醇 /氯仿溶液洗脱。所得到的产物行 1 H- NMR(核磁共振 )和 1 3C- NMR测定。结果发现含甲醇 6 0 %的甲醇 /氯仿溶液的洗脱物为纯化 Glob- N (红细胞糖苷脂 ) ,层析展开后呈单一清晰条带 ,Rf值 0 .45。初步结构分析表明牛脑 Glob- N含 N-乙酰半乳糖 ,共 3个糖基 ,其神经酰胺部分含有 2 6个碳。结果表明柱层析分离中性鞘糖脂单一组分效果好 ,纯度高。牛脑 Glob- N结构中含 3个糖基
In order to explore a new method for the purification, separation and identification of single components of neutral glycosphingolipids (N-GSLs), bovine brain neutral glycosphingolipids extracted by modified Hakomori method were subjected to pressure column chromatography. Gradient methanol/chloroform elution. The resulting product was subjected to 1 H-NMR (NMR) and 13C-NMR measurements. The results showed that the eluate containing 60% methanol in methanol/chloroform solution was purified Glob-N (erythrocyte glycoside), and the chromatogram developed a single clear band with an Rf value of 0.45. Preliminary structural analysis showed that the bovine brain Glob-N contains N-acetylgalactose, a total of 3 glycosyl groups, and its ceramide moiety contains 26 carbons. The results showed that the column chromatography separation of neutral sheath sugar lipid single component effect is good, high purity. The brain contains 3 sugar groups in the Glob-N structure
