目的 制备分泌抗 HBs Ag单克隆抗体杂交瘤细胞株 ,为建立快速诊断 HBs Ag的方法奠定了基础。 方法 通过细胞融合建立了能稳定分泌抗 HBs Ag单抗的杂交瘤细胞株 ,利用 EL ISA技术测定单抗的特异性 ,并用此单抗对临床标本进行初步检测。 结果 细胞株分泌的单抗可与 HBs Ag特异性结合 ,与乙肝病毒 E抗原和核心抗原无交叉反应 ,经免疫扩散试验分析为 Ig G1型 ,将此单抗用辣根过氧化物酶标记后可与 HBs Ag阳性的病人血清反应。 结论 筛选出了一株适合临床应用的单抗杂交瘤细胞株。
OBJECTIVE: To prepare a hybridoma cell line secreting anti-HBs Ag monoclonal antibody, which lays the foundation for the establishment of a rapid method for the diagnosis of HBsAg. Methods The hybridoma cell line that can stably secrete anti-HBs Ag monoclonal antibody was established by cell fusion. The specificity of the McAb was determined by ELISA technique. The clinical samples were preliminarily tested by this monoclonal antibody. Results The monoclonal antibodies secreted by cell lines could specifically bind to HBs Ag and did not cross-react with hepatitis E virus E antigen and core antigen. Ig G1 was detected by immunodiffusion test. The McAb was labeled with horseradish peroxidase Serum reaction with HBs Ag-positive patients. Conclusion A monoclonal antibody hybridoma cell line suitable for clinical application was screened out.
