目的探讨鞘氨醇激酶/1-磷酸鞘氨醇信号途径(SPK/S1P)对人肝癌细胞增殖、凋亡的影响。方法体外培养人肝癌细胞株HepG2,观察5~20μmoL/L浓度范围鞘氨醇激酶的特异性抑制剂N,N′-二甲基鞘氨鞍醇(DMS)对HepG2细胞增殖、凋亡的影响。结果MTT实验结果显示,5~20μmoL/L浓度范围的DMS对细胞增殖均有抑制作用,透射电镜观察对照组细胞可见细胞凋亡的变化,TUNEL标记法检测处理组阳性细胞表达百分数较对照组明显增多(P<0.01)。结论DMS对HepG2细胞的生长增殖具有抑制作用,并促其发生凋亡。
Objective To investigate the effects of sphingosine kinase / sphingosine-1 -phosphate signal pathway (SPK / S1P) on proliferation and apoptosis of human hepatoma cells. Methods Human hepatocellular carcinoma cell line HepG2 was cultured in vitro. The effects of sphingosine kinase specific inhibitor N, N’-dimethylpylsalphaneol (DMS) on the proliferation and apoptosis of HepG2 cells were observed at 5 ~ 20μmoL / L . Results MTT assay showed that DMS concentration in the range of 5 ~ 20μmoL / L inhibited the proliferation of cells. The changes of apoptotic cells were observed by transmission electron microscopy. The percentage of positive cells detected by TUNEL assay was significantly higher than that of the control group Increased (P <0.01). Conclusion DMS can inhibit HepG2 cells proliferation and induce apoptosis.
