目的 探讨 1,2 -二氯乙烷 (1,2 -DCE)对人外周血淋巴细胞DNA的损伤作用及胆红素的保护作用。方法 用不同浓度的 1,2 -DCE对人外周血淋巴细胞进行染毒 ,然后用单细胞凝胶电泳法观察DNA链断裂、DNA -DNA链间交联和DNA损伤后的修复能力 ,同时观察胆红素对 1,2 -DCE所致DNA损伤的保护作用。结果 5 0~ 2 5 0 μmol/L的 1,2 -DCE均可明显引起淋巴细胞DNA的链断裂 ;5 0 ,10 0 ,2 0 0 μmol/L的 1,2 -DCE还可引起淋巴细胞DNA -DNA链间交联 ,其交联率分别为 37 9% ,42 8%和 48 5 % ;2 5 0 μmol/L的 1,2 -DCE还可明显抑制淋巴细胞DNA损伤后的修复能力 ;10或 2 0 μmol/L胆红素可明显抑制 1,2 -DCE所致的淋巴细胞DNA链断裂和DNA -DNA链间交联 ,并可加速 1,2 -DCE所致的淋巴细胞DNA损伤后的修复。结论 1,2 -DCE可引起淋巴细胞DNA的多种损伤 ,胆红素对这些损伤具有保护作用
Objective To investigate the DNA damaging effect of 1,2-dichloroethane (1,2-DCE) on human peripheral blood lymphocytes and the protective effect of bilirubin. Methods Human peripheral blood lymphocytes were exposed to different concentrations of 1,2-DCE. Single-cell gel electrophoresis was used to observe DNA strand breaks, DNA-DNA strand cross-links, and DNA repair capacity. Protective Effect of Bilirubin on DNA Damage Induced by 1,2-DCE. RESULTS: 1,2, -DCE at concentrations ranging from 50 to 250 μmol / L could significantly induce DNA strand breaks in lymphocytes. Both 50 and 100 micromolar concentrations of 1,2-DCE at 200 μmol / L could induce lymphocyte The cross-linking of DNA-DNA chains was 37 9%, 42 8% and 48 5%, respectively. 1,2-DCE at 250 μmol / L also significantly inhibited the repair ability of DNA after DNA damage ; 10 or 20 μmol / L bilirubin significantly inhibited 1,2-DCE-induced lymphocyte DNA strand breaks and DNA-DNA strand cross-links and accelerated 1,2-DCE-induced lymphocyte DNA Repair after injury. Conclusion 1,2-DCE can cause various damage of lymphocyte DNA, and bilirubin has a protective effect on these injuries
