背景:补肾法正逐步成为中医老年病学中抗衰老的治疗方法之一。目的:探讨滋补肾阴方左归饮对老年小鼠自由基代谢血清丙二醛含量,超氧化物歧化酶活性的影响。设计:完全随机对照实验。单位:广东医学院微生物与免疫学教研室。对象:实验于2002-09/2004-07在皖南医学院免疫学研究室完成,选择四川省医学科学院实验动物研究所提供的40只雄性老年封闭群小鼠。随机分为老年对照组、左归饮组、单味熟地组及去熟地组,每组10只。左归饮由熟地、山药、枸杞、炙甘草、茯苓、山茱萸,按9:6:6:3:4:5比例组成。常规水煎,左归饮组浓缩至含生药0.5775g/mL,单味熟地组浓缩至含生药0.1575g/mL,去熟地组浓缩至含生药0.42g/mL,配成100g/L浓度的药水混合液。干预:左归饮组、单味熟地组、去熟地组饮用左归饮、熟地、去熟地,4次/d,1.5mL/次,共75d,给药期间不提供其他饮用水。老年对照组小鼠饮用自来水。饮用75d后,摘眼球取血2mL,留存血清,用硫代巴比妥酸法测定血清中丙二醛含量,黄嘌呤氧化酶法测定血清中超氧化物歧化酶活性,比色法测定血清总抗氧化能力。主要观察指标:①血清中丙二醛含量。②血清中超氧化物歧化酶活性。③血清总抗氧化能力检测。结果:老年对照组死亡1只,单味熟地组死亡3只,最终36只小鼠进入结果分析。①血清中丙二醛含量:左归饮组和单味熟地组低于老年对照组[(5.280±1.204)μmol/L和(5.886±2.717)μmol/L,(9.533±3.494)μmol/L,(q=3.641~4.657,P<0.05)]。左归饮组和单味熟地组老年小鼠血清中丙二醛含量基本一致。②超氧化物歧化酶活性:左归饮组和去熟地组高于老年对照组[(7.008±0.782)mkat/L和(6.989±0.809)mkat/L,(5.908±0.392)mkat/L,(q=3.641~4.657,P<0.05)],左归饮组和去熟地组超氧化物歧化酶活性基本一致。单味熟地组与老年对照组超氧化物歧化酶活性基本一致。③总抗氧化能力:左归饮组和单味熟地组虽然高于老年对照组,去熟地组低于老年对照组,但差异均无显著性意义(P>0.05)。结论:左归饮能明显降低老年小鼠血清中丙二醛含量,并能提高血清中超氧化物歧化酶活性,表明左归饮在改善老年机体自由基代谢、增强超氧化物歧化酶活性和减少丙二醛积蓄对机体损伤方面具有明显作用。
Background: The kidney-building method is gradually becoming one of the anti-aging treatment methods in the geriatrics of TCM. Objective: To investigate the effect of Zigui Kidney Decoction Zuoguiyin on serum malondialdehyde (MDA) and superoxide dismutase (SOD) activity in aged mice. Design: Completely randomized controlled trials. Unit: Department of Microbiology and Immunology, Guangdong Medical College. PARTICIPANTS: The experiment was performed at the Laboratory of Immunology, Wannan Medical College from September 2002 to July 2004. A total of 40 male closed-group mice were selected from Laboratory Animal Institute of Sichuan Academy of Medical Sciences. They were randomly divided into old control group, Zuoguiyin group, single-flavored rehmannia group and decoction group, 10 in each group. Zuoguiyin consists of Rehmannia, yam, wolfberry, licorice root, alfalfa, and hawthorn, and is composed of 9:6:6:3:4:5 ratios. In conventional decoction, Zuoguiyin group was concentrated to 0.5775g/mL crude drug, single-taste rehmannia group was concentrated to 0.1575g/mL crude drug, and decoction group was concentrated to 0.42g/mL crude drug, and 100g/L concentration of syrup was prepared. Mixture. Intervention: Zuoguiyin group, single-flavored rehmannia group, and decoction group were treated with Zuoguiyin, rehmannia, and decoction, 4 times/d, 1.5mL/time for 75 days, and no other drinking water was provided during the administration. Older control mice were given tap water. After 75 days of drinking, 2 mL of blood was taken from the eyeball, serum was retained, the content of malondialdehyde in the serum was measured by thiobarbituric acid method, the activity of superoxide dismutase in the serum was measured by the xanthine oxidase method, and the serum total resistance was determined by colorimetric method. Oxidation capacity. MAIN OUTCOME MEASURES: 1 Serum malondialdehyde content. 2 Superoxide dismutase activity in serum. 3 serum total antioxidant capacity test. RESULTS: There were 1 dead in the old control group and 3 in the single-flavored rehmannia group. The final 36 mice entered the result analysis. 1 Malondialdehyde content in serum: Compared with the old control group, [(0.280±1.204) μmol/L and (5.886±2.717) μmol/L, (9.533±3.494) μmol/L] in the Zuoguiyin group and the single-flavoured rehmannia group, respectively. (q=3.641 to 4.657, P<0.05)]. The content of malondialdehyde in serum of elderly mice of Zuoguiyin group and single-flavored rehmannia group was basically the same. 2 Superoxide dismutase activity: the Zuoguiyin group and the decoction group were higher than the old control group [(7.008±0.782)mkat/L and (6.989±0.809)mkat/L, (5.908±0.392)mkat/L, ( q=3.641~4.657, P<0.05)]. The activities of superoxide dismutase in Zuoguiyin group and the decoction group were basically the same. The superoxide dismutase activity of the single-flavored rehmannia group was similar to that of the old-aged control group. 3 Total antioxidant capacity: Although the left and right rehmannia groups were higher than the old rehmannia group, the decoction group was lower than the old group, but the difference was not significant (P>0.05). Conclusion: Zuoguiyin can significantly reduce the content of malondialdehyde in serum of aged mice and increase the activity of superoxide dismutase in serum. It is indicated that Zuoguiyin improves free radical metabolism and enhances the activity of superoxide dismutase in old people. Malondialdehyde accumulation has a significant effect on body damage.
