2,4-滴丁酯对雄性小鼠睾丸的氧化损伤及血清睾酮的影响

[目的]探讨2,4-滴丁酯对雄性小鼠睾丸的氧化损伤及血清睾酮的影响。[方法]将48只清洁级健康雄性ICR小鼠按体重随机分为4组,每组12只,分别为对照组(玉米油)和低(10 mg/kg)、中(20 mg/kg)、高剂量(40 mg/kg)2,4-滴丁酯染毒组。采用灌胃的方式进行染毒,1次/d,每周连续染毒6 d,实验周期为5周。染毒结束24 h后,采用放射免疫分析法测定血清中睾酮的含量;取睾丸测定脏器系数及采用分光光度法对睾丸组织中超氧化物歧化酶(SOD)活力和丙二醛(MDA)的含量进行测定。[结果]染毒4周时,与对照组比较,中剂量组和高剂量组小鼠体重增长幅度明显降低,差异有统计学意义(P<0.05);染毒5周时,与对照组和低剂量组相比,中剂量组与高剂量组小鼠体重增长幅度明显降低,差异有统计学意义(P<0.05);小鼠睾丸脏器系数随染毒浓度的增大呈下降趋势,与对照组和低剂量组相比,高剂量组睾丸脏器系数值较低,且差异有统计学意义(P<0.05);SOD活力随着2,4-滴丁酯染毒剂量的升高逐渐降低,各实验组间差异均有统计学意义(P<0.05);MDA含量随着2,4-滴丁酯染毒剂量的升高逐渐增高,高剂量组与对照组之间的差异有统计学意义(P<0.05);血清中睾酮水平随着染毒剂量的升高逐渐降低,与对照组和低剂量组比较,高、中剂量组血清中睾酮水平较低,差异有统计学意义(P<0.05),且高剂量组与中剂量组之间差异有统计学意义(P<0.05)。[结论]2,4-滴丁酯可导致雄性小鼠睾丸组织氧化损伤并降低血清中睾酮水平。 [Objective] To investigate the effects of 2,4-D butyl ester on oxidative damage of testis and serum testosterone in male mice. [Methods] Forty-eight healthy male ICR mice of clean grade were randomly divided into 4 groups according to body weight, 12 in each group, which were control group (corn oil) and low (10 mg / kg) , High dose (40 mg / kg) 2,4-D butyl ester exposure group. The rats were treated by gavage once a day for 6 days. The experiment period was 5 weeks. Serum testosterone levels were measured by radioimmunoassay 24 hours after the end of treatment. The organ coefficient of testis was measured and the activity of superoxide dismutase (SOD) and malondialdehyde (MDA) in testis tissue were determined by spectrophotometry Content was measured. [Result] Compared with the control group, the body weight gain of the middle-dose and high-dose groups was significantly decreased at 4 weeks after exposure (P <0.05). At 5 weeks of exposure, compared with the control group and Compared with the low-dose group, the body weight gain of the middle-dose and high-dose groups was significantly lower (P <0.05); the testis organ coefficient of the mice showed a downward trend with the increase of the concentration, Compared with the low-dose group, the testis organ coefficient of the high-dose group was lower, and the difference was statistically significant (P <0.05); SOD activity gradually increased with the increase of 2,4- (P <0.05). The content of MDA increased gradually with the increase of the dose of 2,4-Dbutyltransferase, and the difference between the high-dose group and the control group was statistically significant (P <0.05). The level of testosterone in serum decreased gradually with the increase of exposure dose. Compared with control group and low dose group, serum testosterone level was lower in high dose group and middle dose group, with significant difference (P < P <0.05), and the difference between the high-dose group and the medium-dose group was statistically significant (P <0.05). [Conclusion] 2,4-D butyl ester can lead to the oxidative damage of testicular tissue and the serum testosterone level in male mice.
