用体外培养的骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMMSCs)治疗深Ⅱ度烫伤大鼠。体外培养雄性Wistar大鼠BMMSCs,流式细胞仪鉴定。将BMMSCs分别通过创面局部、尾静脉注射移植给深Ⅱ度烫伤的雌性大鼠。观察创面愈合情况;不同时间点取组织标本制作石蜡切片,HE染色观察;PCR检测创面皮肤组织和骨髓中供体鼠Y染色体基因性别决定区(Sry)片段的表达。结果显示,静脉注射组和创面局部注射组愈合速度均快于对照组(P<0.05),但两治疗组之间比较差异无显著性(P>0.05)。HE染色结果显示治疗组创面愈合质量均好于对照组。两治疗组动物创面组织及静脉注射组骨髓组织均有Y染色体基因的表达。因此,大鼠供体骨髓MSCs对其受体皮肤深Ⅱ度烫伤具有一定的治疗作用。烫伤促使BMMSCs向创面迁移并参与烫伤创面愈合。
Bone marrow mesenchymal stem cells (BMMSCs) cultured in vitro were used to treat deep second degree scalded rats. The cultured male Wistar rats BMMSCs were identified by flow cytometry. The BMMSCs were transplanted into the deep Ⅱ degree scalded female rats through tail wound and tail vein respectively. The healing of wounds was observed. Paraffin sections were made at different time points and stained with hematoxylin-eosin (HE). The expression of Y chromosome Sry was detected by PCR in skin and bone marrow. The results showed that the healing rate of injection group and wound injection group were faster than that of control group (P <0.05), but there was no significant difference between the two treatment groups (P> 0.05). The results of HE staining showed that the wound healing quality of the treatment group was better than that of the control group. The expression of Y chromosome gene was detected in both wounds and intravenous injection groups. Therefore, the rat donor bone marrow MSCs have a certain therapeutic effect on deep Ⅱ degree scald skin. Scald prompted BMMSCs to migrate to the wounds and participate in burn wound healing.
