目的评价硕苞蔷薇提取物RBa -I对抗吗啡 (Mor)依赖大鼠戒断症状的作用 ,并初步探讨其机理。方法造成Mor依赖大鼠的模型 ,然后考察RBa -I对抗Mor戒断症状的作用及其对下丘脑和垂体中 β -内啡肽 (β-EP)含量的影响。结果硕苞蔷薇提取物RBa-I (5mg·kg- 1,ig)能对抗Mor依赖大鼠催促戒断症状 ,使总积分显著减少 ,但作用比可乐定弱 ;能对抗Mor依赖大鼠自然戒断症状 ,且能对抗Mor依赖大鼠体重减轻 ;经过 7d治疗以后 ,能明显控制戒断症状 ;升高Mor依赖大鼠下丘脑和垂体中 β-EP的含量。结论硕苞蔷薇提取物RBa -I能对抗Mor依赖大鼠戒断症状 ,使下丘脑和垂体中 β -EP的含量升高。
OBJECTIVE: To evaluate the effect of the extract of Rhizoma Osmanthus Rba-I against morphine (Mor)-dependent withdrawal symptoms in rats and to explore its mechanism. METHODS: The Mor-dependent rat model was used to investigate the effect of RBa-I against Mor withdrawal symptoms and its effect on β-endorphin (β-EP) levels in the hypothalamus and pituitary. Results The extract of R. rosenbergii Rba-I (5mg·kg-1, ig) can relieve withdrawal symptoms in Mor dependent rats and significantly reduce the total score, but the effect is weaker than that of clonidine; it can protect against natural growth in Mor-dependent rats. Symptoms were broken, and they were able to counteract the weight loss of Mor-dependent rats; after 7 days of treatment, the withdrawal symptoms were significantly controlled; and the β-EP levels in the hypothalamus and pituitary of Mor-dependent rats were increased. Conclusion The extract of R. rosenbergii Rba-I can resist the withdrawal symptoms in Mor-dependent rats, and increase the content of β-EP in the hypothalamus and pituitary.
