目的 :研究mPEG有效修饰血小板表面HLA抗原的方法及效果。方法 :在 2 2℃、pH 7.4的PBS介质中 ,采用浓度梯度法修饰血小板表面的HLA抗原。并分别用处理前后的血小板吸收血清中的相应抗体 ,通过微量混合淋巴细胞毒试验检测化学修饰的效果。结果 :经mPEG处理后的血小板无吸收血清中相应抗体的能力 ,微量淋巴细胞毒试验为阳性 ;未经mPEG处理的血小板则与之相反。结论 :血小板表面的HLA抗原经mPEG修饰后 ,可完全阻断其与相应抗体间的特异性免疫反应
Objective: To study the method and effect of mPEG on effectively modifying HLA antigens on platelet surface. Methods: HLA antigens on platelets were modified by concentration gradient method in PBS at pH 7.4. The corresponding antibodies in the serum before and after platelet absorption were tested respectively, and the effect of chemical modification was detected by micromix lymphocytotoxicity test. Results: The mPEG-treated platelets did not have the ability to absorb the corresponding antibodies in the serum, and the microtiter cytotoxicity test was positive. The platelets without mPEG treatment were the opposite. Conclusion: The platelet surface of HLA antigens modified by mPEG can completely block the specific immune response with the corresponding antibodies
