目的:探讨在1%氧浓度下及6%氧浓度下体外培养正常妊娠孕妇胎盘绒毛组织培养液中代谢足迹的变化。方法:选择正常妊娠孕妇27例,在胎盘边缘、中间、近脐带处取绒毛组织,分别在1%氧浓度下及6%氧浓度下培养96 h,采用高效液相色谱-质谱法检测培养液中代谢产物,观察不同氧浓度下培养的绒毛组织代谢足迹的变化。结果:1%氧浓度和6%氧浓度下分别取自同一胎盘边缘、中间、近脐带三点处胎盘绒毛组织代谢足迹比较,无统计学差异(P>0.05);1%氧浓度与6%氧浓度下培养的孕妇胎盘绒毛组织代谢产物比较,发现545个代谢物有统计学差异(P<0.05)。结论:代谢组学提供了从另一个角度研究胎盘功能的方法,应用严谨的实验技术和适当的统计学分析,能确定体外培养的胎盘组织在不同氧浓度下的代谢差异,从而研究与胎盘病理学相关的改变。
OBJECTIVE: To investigate the changes of metabolic footprint in placental chorionic villi in normal pregnant women incubated with 1% oxygen and 6% oxygen. Methods: Twenty-seven pregnant women of normal pregnancy were selected, and the villi were taken from the marginal, middle and proximal umbilical cord of the placenta, respectively. The culture was carried out at 1% oxygen concentration and 6% oxygen concentration for 96 h, respectively. The culture medium was detected by high performance liquid chromatography In metabolites, to observe the changes of metabolic footprint of villi cultured under different oxygen concentrations. Results: There was no significant difference in the metabolic footprint of placental villi between the same placenta at 1% oxygen concentration and 6% oxygen concentration (P> 0.05), while the difference between 1% oxygen concentration and 6% Metabolites of placental villi in pregnant women incubated at oxygen concentration showed 545 metabolites were statistically different (P <0.05). CONCLUSION: Metabolomics provides a method of studying placental function from another perspective. Using rigorous experimental techniques and appropriate statistical analyzes to determine the metabolic differences in placental tissue cultured at different oxygen concentrations in vitro, Neo-Confucianism-related changes.
