目的研究骨桥蛋白在HCC中的表达与临床病理特征的关系。方法选取48例HCC手术切除的新鲜肝癌和癌旁组织标本和10例正常肝组织,以10%福尔马林固定,常规石蜡包埋,每一个标本行连续切片,厚4μm。应用SABC免疫组化法,以骨桥蛋白单克隆抗体对HCC组织切片进行染色。对骨桥蛋白在HCC中的表达及其临床病理特征的关系进行分析。结果OPN在HCC组织中的阳性率为50.00%,明显高于癌旁及正常肝组织(P<0.01)。骨桥蛋白在高、中、低HCC中评分依次为0.67±1.63,2.14±1.81和2.20±1.74,低分化明显高于高分化(P<0.05)。骨桥蛋白在有转移与无转移HCC中的评分是2.64±1.84和1.25±1.74,有转移者明显高于无转移者(P<0.01)。骨桥蛋白与其他病理特征无明显关系。结论骨桥蛋白在HCC癌组织中的表达明显升高,低中分化者和伴有转移者高于分化高和不伴转移者。提示骨桥蛋白可能促进了HCC的发展和转移。
Objective To study the relationship between osteopontin expression and clinicopathological features in HCC. Methods 48 cases of HCC surgically resected fresh hepatocellular carcinoma and para-cancerous tissue specimens and 10 normal liver tissues were selected and fixed in 10% formalin and paraffin-embedded. Each specimen was serially sectioned and the thickness was 4 μm. SABC immunohistochemistry was used to stain HCC tissue sections with osteopontin monoclonal antibody. The relationship between osteopontin expression in HCC and its clinicopathological features was analyzed. Results The positive rate of OPN in HCC tissues was 50.00%, which was significantly higher than that in adjacent normal tissues and normal liver tissues (P <0.01). Osteopontin scores were 0.67 ± 1.63, 2.14 ± 1.81 and 2.20 ± 1.74 in high, medium and low HCC, respectively. The low differentiation was significantly higher than that in well-differentiated (P <0.05). The osteopontin score was 2.64 ± 1.84 and 1.25 ± 1.74 in metastatic and non-metastatic HCC patients, with significantly higher metastasis than those without metastasis (P <0.01). Osteopontin has no obvious relationship with other pathological features. Conclusion The expression of osteopontin in HCC tissues was significantly higher than those in poorly differentiated and metastatic cases. Tip osteopontin may promote the development and metastasis of HCC.
