目的:研究葡萄糖激酶(GCK)基因多态性与2型糖尿病的关系,并获得GCK基因频率在中国人群中的分布规律。方法:运用聚合酶联反应技术结合短串联重复序列(STR)的方法检测107例2型糖尿病患者及101例正常对照者的GCK基因型。结果:两组1、2、3、4和5等位基因的分布频率分别为0.08、0.46、0.07、0.29、0.10和0.11、0.49、0.16、0.23、0.01,其总体频率分布存在显著性差异,2型糖尿病组的5等位基因频率明显高于对照组。比较GCK基因座在中国人群与欧洲人及北美人群的分布,发现存在明显的种族差异。结论:GCK基因多态标记与中国人2型糖尿病相关联。
Objective: To investigate the relationship between glucokinase (GCK) gene polymorphism and type 2 diabetes mellitus (T2DM) and to obtain the distribution of GCK gene frequency in Chinese population. Methods: GCK genotypes of 107 patients with type 2 diabetes mellitus and 101 normal controls were detected by polymerase chain reaction and short tandem repeat (STR). Results: The frequencies of alleles 1, 2, 3, 4 and 5 in two groups were 0.08,0.46,0.07,0.29,0.10 and 0.11,0.49,0.16,0.23,0.01, respectively The overall frequency distribution was significantly different, type 2 diabetes group 5 allele frequency was significantly higher than the control group. Comparing the distribution of GCK loci in Chinese and Europeans and North Americans, we found significant racial differences. Conclusion: GCK polymorphism markers are associated with type 2 diabetes in Chinese.
