为了优化替考游动放线菌FIM-37菌株产替考拉宁发酵条件,采用单因素试验和正交试验研究发酵培养基组成,探讨摇瓶发酵的主要影响因子初始pH值、装液量、转速、温度等发酵参数的影响.结果确定了适宜发酵培养基和发酵参数:可溶性淀粉3.0%,葡萄糖1.0%,黄豆粉4.5%,蛋白胨0.5%,磷酸氢二钾0.02%,氯化镁0.05%,硫酸铵0.02%,脯氨酸0.1%,碳酸钙0.3%,初始pH7.5 ~8.0,装液量50 mL/500 mL,种龄40~48 h,接种量5%,摇床转速230 r/min,发酵温度30℃,发酵时间8d.优化后的发酵水平较原工艺提高了40%以上,该优化发酵工艺为替考拉宁的工业化生产奠定了基础.","To optimize the fermentation conditions of teicoplanin producing strain A.teichomyceticus FIM-37,the single factor experiments and orthogonal experiments were conducted to study the liquid fermentation medium and parameters of A.teichomyceticus FIM-37 fermentation.It was determined that the appropriate fermentation medium formula composition and optimal fermentation parameters were as follows:soluble starch 3.0%,glucose 1.0%,soybean flour 4.5 %,peptone 0.5 %,K2HPO40.02%,and MgCl2 0.05 %,(NH4)2SO4 0.02%,proline 0.1%,CaCO3 0.3 %,initial pH at 7.5 ~ 8.0,filling volume 50 mL for a 500 mL shake flask,cultivation time of seed 40 ~48 h,inoculum size 5%,shaker's rotating speed 230 r/min,fermentation temperature 30 ℃,and fermentation time 8 d.The teicoplanin yield of A.teichomyceticus FIM-37 on the optimal fermentation conditions was enhanced more than 40%higher than that of the original.It laid a foundation for producing teicoplanin on industrial scale.
