目的:建立同时测定五味子果实中10种木脂素类成分含量的方法,分析木脂素类成分在五味子果实的种仁、种皮和果肉中的含量差异,探索各成分的分布规律。方法:采用高效液相色谱法(HPLC)测定不同产地五味子果实中五味子醇甲,戈米辛D,戈米辛J,五味子醇乙,当归酰戈米辛H,戈米辛G,五味子酯甲,五味子甲素,五味子乙素,五味子丙素的含量,流动相乙腈(A)-水(B)梯度洗脱(0~30 min,40%A;30~32 min,40%~46%A;32~53 min,46%A;53~64 min,46%~50%A;64~83 min,50%~57%A;83~107 min,57%~62%A;107~115 min,62%~100%A;115~120 min,100%A),检测波长220nm,进样量4μL。结果:不同产地五味子果实的种仁、种皮和果肉中总木脂素质量分数分别为30.08,9.76,2.24 mg·g-1。10种木脂素类成分在五味子果实3个不同部位中的含量差异极显著,排序为种仁>种皮>果肉。结论:建立的含量测定方法能较好地反映五味子果实的不同部位中木脂素类成分的分布信息,为指导五味子果实的临床合理应用提供参考。
OBJECTIVE: To establish a method for simultaneous determination of 10 kinds of lignans in the fruit of Schisandra chinensis fruit, and to analyze the content differences of lignan in the seed kernel, seed coat and pulp of Schisandra chinensis fruit and to explore the distribution rules of each component. Methods: The content of schisandrin A, Gomisin D, Gomisin J, Schizandrol B, angelica gomisin H, Gomisin G, schisandrin A , Schisandrin A, Schisandrin B and Schisandra C, and the mobile phase was eluted with a gradient of acetonitrile-water (B) (0-30 min, 40% A; 30-32 min, 40-46% A ; 32-53 min, 46% A; 53-64 min, 46% -50% A; 64-83 min, 50% -57% A; 83-107 min, 57% -62% A; , 62% ~ 100% A; 115 ~ 120 min, 100% A), detection wavelength 220nm, injection volume 4μL. Results: The total lignan contents in seeds, seed coat and pulp of Fructus schisandrae from different areas were 30.08, 9.76 and 2.24 mg · g-1.10 kinds of lignan components were found in three different parts of Schisandra chinensis fruit There was significant difference in content between seeds, seed coat and pulp. Conclusion: The established method for determination of content can reflect the distribution information of lignans in different parts of Schisandra fruit well and provide a reference for clinical application of Schisandra sphenanthera fruit.
